This paper aimed to study on the therapeutic effect of Sauropus rostratus Miq.(SRMID.)and Jaceosidin on lipopolysaccharide-induced acute lung injury(ALI)in mice.The therapeutic effect mechanisms of Jaceosidin was explored in antiinflammatory,complement and oxidative stressthree pathways.The therapeutic effect mechanisms of Sauropus rostratus Miq.Was decreased proinflammatory cytokines,up-regulation of antiinflammatory factors,and antioxidant stress.PartⅠThe therapeutic effect of Jaceosidin on lipopolysaccharide-induced acute lung injury in miceJaceosidin extracted by the research group from the medicinal herbs isolated in a flavonoids(RJFs).With anti-tumor,immunosuppression and anti-insulin resistance to diabetes and other activities.retreatment with RJFs could significantl decrese lung(W/D)ratio,nitric oxide(NO)and Protein concentration in BALF,which was exhibited together with the lowered MPO activity,the increased suPeroxide dismutase(SOD)activity and down-regulation the level of TNF-α,IL-6 and I-1β in ALI model.Further study found pretreatment with Jaceosidin could significantly decrease lung W/D ratio,Protein concentration in BALF,which was exhibited together with the lowered myeloperoxidase(MPO)activity,the increased CAT activity,down-regulation the level of TNF-α,IL-6 and IL-1β and increased the level of IL-10 and IL-4 in ALI model.Additionally,Jaceosidin attenuated lung histopathological changes and significantly reduced NF-κB 、COX-2 and complement3(C3)dePosition.Furthermore,Jaceosidin decreased the level of NF-κB 、COX-2 and C3 in serum.PartⅡ The therapeutic effect of Sauropus spatulifolius Beille on lipopolysaccharide-induced acute lung injury in miceSauropus spatulifolius Beille is used for treatment of cough,Catharsis and Sore throat.Anti-complementary activity test in vitro indicated that the water extract of S.spatulifolius Beille has the effect of treating acute lung injury.Pretreatment with LLY40 could significantly decrease lung wet-to-dry weight(W/D)ratio,protein concentration in BALF,which was exhibited together with the lowered myeloperoxidase(MPO)activity,the increased superoxide dismutase(SOD)and Catalase(CAT)activity and down-regulation the level of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β)in ALI model.Additionally,LLY40 attenuated lung histopathological changes.9 compounds were isolated from LLY40,their structures were identified by their physico-chemical charateristics and detailed spectroscopic analysis.The compounds were identified as Aurantiamide,Aurantiamideacetate,(-)-lyoniresinol,(-)-lyoniresinol 3α-Oβ-Dglucopyranoside.Catechol,(-)-lyoniresinol,(-)-lyoniresinol 3α-Oβ-D-glucopyranoside,aurantiamide is isolated from the Sauropus rostratus Beille for the first time.Determination of polyphenol content by fulin phenol reagentmethod,LLY40 polyphenol content of 24.3%.In summary,The therapeutic effect mechanisms of Jaceosidin was Related with NF-κB(Blocking the regulation of inflammatory factors in its synthesis),COX-2 and complement three pathways.LLY40 has a therapeutic effect on LPS-induced ALI in mice,which is associated with decreased Proinflammatory cytokines,up-regulation of antiinflammatory factors,and antioxidant stress.
