Objective To investigate the difference of the expression of Beclin1 and PTEN in colorectal cancer tissue and paracancerous tissues and their correlation with prognosis of colorectal cancer.Methods Collect the colorectal cancer tissue,para cancerous tissues and clinical data of 127 patients with colorectal cancer who were diagnosed and operated in The 94 th Hospital of Chinese People’s Liberation Army from 2007 to 2017.Real-time PCR was used to examine the mRNA expression of Beclin1 and PTEN in these tissues,meanwhile Western-blot was used to examine the protein expression of these tissues.Linear correlation was used to analyzes the relationship between the protein expression of Beclin1 and PTEN,futher more research the correlation between expression of Beclin1 and PTEN with colorectal cancer prognosis.Results Beclin1 localized in the cytoplasm of colorectal cells,whereas PTEN is localized in the nucleus.Beclin1 mRNA and protein expression in cancer tissue was significantly higher than paracancerous tissues,however PTEN was significantly lower(P<0.01).Linear correlation showed that Beclin1 protein expression was negative correlation with PTEN(P<0.01,R2=0.223).The positive expression rates of Beclin1 and PTEN were related to the degree of differentiation,depth of invasion,lymph node metastasis,and TNM staging,respectively.The five-year survival rate of Beclin1 and PTEN protein positive patients was significantly higher than that of negative patients;the survival of patients with single factor analysis In addition,the degree of tumor differentiation,depth of invasion,lymph node metastasis,distant metastasis,TNM stage,and expression of Beclin 1 and PTEN protein were all related to the survival of patients.Cox regression multivariate analysis suggested that whether distant metastasis,lymph node metastasis,and Beclin 1 and PTEN protein expression were independent factors in judging the survival time of patients with colorectal cancer,and Beclin 1 and PTEN proteins were protective factors.The survival rate of patients with positive expression of both proteins was significantly higher than that of double negative patients.Conclusions Beclin1 and PTEN may participate in the pathogenesis of colorectal cancer and may predict their prognosis.
