Objective:The effects of rifampicin combined with ambroxol hydrochloride on the biofilm formation of Staphylococcus epidermidis were investigated by observing the effects of rifampicin and ambroxol hydrochloride on the biofilm formation of Staphylococcus epidermidis in orthopedic implants.This was the basis of the experiment.Methods:1.The effect of ambroxol hydrochloride on biofilm formation on different rough materials was investigated by in vitro experiment.The effects of Staphylococcus epidermidis on the surface adhesion of titanium and polyethylene were observed by fluorescent staining.The effects of different materials and the roughness of artificial materials on the adhesion of Staphylococcus epidermis were observed under the action of ambroxol hydrochloride.Finally,the effect of ambroxol hydrochloride on the surface adhesion and formation of Staphylococcus epidermidis biofilm in orthopedic implants was studied by using SYT09 to compare the difference between live bacteria and PI in biofilm.2.The effect of rifampicin combined with ambroxol hydrochloride on theformation of bacterial biofilm was studied by animal experimentA model of artificial prosthesis infection was established in New Zealand rabbits.Three weeks later,rifampicin and ambroxol were injected into the knee joint of the inoculated bacteria to observe the general condition of the rabbits,the white blood cell count and the biofilm implanted into the prosthesis.The number of bacteria in the infected model was cultured in the tissues around the knee,and the morphology of the bacterial biofilm was observed by fluorescence microscopy and laser confocal microscopy.Results:1.The effect of different types of material affects the adhesion of Staphylococcus epidermidis to the biofilm;ambroxol can inhibit the adhesion of Staphylococcus epidermidis biofilm,rifampicin combined with Ambroxol hydrochloride can enhance the inhibition of bacterial biofilm formation.2.Ampiciosone hydrochloride is effective in the prevention and treatment of prosthesis infection after artificial knee prosthesis implantation.Ambroxol hydrochloride can increase the bactericidal effect of rifampicin.Conclusion:1.Ambroxol hydrochloride can inhibit the formation of Staphylococcus epidermidis on biofilm,but can not kill it.When combined with rifampicin,it can give more effective control of staphylococcal biofilm formation and kill bacteria.2.Animal experiments show that ambroxol and rifampicin in combination have a significant synergistic effect on Staphylococcus epidermidis bacterial biofilm.
