Purpose:This study aims to investigate the molecular mechanism that TRPM7 can induces the activity of EMT in ovarian cancer cells through calcium signaling.Thereby contributing to the metastasis and invasion of ovarian cancer cells.Methods:The expression of TRPM7 was down-regulated in the cell line sknov3 by transfection with siRNA,changes of intracellular EMT-related markers and transcription factors induced by TRPM7 were detected by Western blot and RT-PCR.Intracellular calcium concentration was decreased by calcium chelator and EMT in cells were induced by EGF,changes of intracellular EMT-related markers and transcription factors absence of calcium concentration were detected by Western blot and RT-PCR,changes in cell migration and invasion were detected by transwell.Down regulating the expression of TRPM7 in skvo3 and intracellular calcium chelator was treated for decreasing the calcium concentration,then EGF was given for inducing EMT in cells,changes of intracellular EMT-related markers and transcription factors absence of calcium ion were detected by Western blot and RT-PCR,changes in cell migration and invasion were detected by transwell.Result:1.Silencing TRPM7 expression led to vimentin,the EGF-induced EMT-related interstitial markers,decreased,and at the same time expressions EMT-related transcription factors,twist and snail,decreased.2.Absence of intracellular calcium led to the significant decrease of EMT-related interstitial markers,vimentin,expressions of EMT-related transcriptional factors twist and snail decreased significantly,abilities of cell migration and invasion decreased significantly(P<0.01).3.Under the condition of normal intracellular calcium,the expression of EMT-related interstitial marker,vimentin in cells with lower expressions of TRPM7 was lower than that of the normal cells,the difference was statistically significant(P<0.01);while under the condition of absence of calcium ion,there was no significant difference in the expression of EMT-related interstitial marker,vimentin between cells with lower expressions of TRPM7 and normal cells(P>0.05),the similar results occurred in the experiments of detecting expressions of EMT-related transcription factors,twist and snail as well as cell migration and invasion.Conclusion:1.TRPM7 may be involved in EGF induced ovarian cancer cell EMT process。2.Calcium signaling may be involved in EGF induced ovarian cancer cell line EMT and promote the migration and invasion of ovarian cancer cells.。3.TRPM7 may mediate EGF induced ovarian cancer cell line EMT by calcium signaling and promote the migration and invasion of ovarian cancer cells。...
