| Purpose:To investigate the effects of aerobic exercise combined with tea polyphenols on oxidative stress and inflammatory cytokines in liver of type 2 diabetic rats,so as to provide experimental basis for prevention and treatment of diabetes mellitus.Experimental method:1)clean SD rats were divided into C group,control group(n = 60),group and diabetic model group(D),with only 52 rats in each group.After one week feeding of common feed,D group was fed with high fat diet for 4 weeks,they was injected intraperitoneally(STZ,30mg ·kg-1· bw-1).The random blood glucose measured at 3d and 7d after injection was 16.7mmol · L-1 and above.2)then the rats were randomly divided into successful type 2 diabetes mellitus group(group DC,8 rats),type 2 diabetes mellitus + exercise group(group DE,8 rats),"type 2 diabetes + TP group(group DT,8 rats),type 2 diabetes mellitus + Exercise + tea polyphenol group(DET group,8).The rats in the exercise group were given no weight-bearing swimming every day for 1 hour and 6 days a week.The tea polyphenols group was treated with the dose of 600mg · kg-1 · d-1 for 8 weeks.After 36 hours ofintervention,rats in each group were fasted overnight,no water,body weight and fasting blood glucose,abdominal anesthesia,abdominal aorta blood and liver tissue.3)Serum levels of insulin,C-reactive protein(CRP),activity of superoxide dismutase(SOD),malondialdehyde(MDA)and IL-6 in liver tissue,and levels of IL-6 and IL-1βmRNA in liver Expression level.Result:1)STZ made into models of diabetes mellitus was 62%,model group symptoms significantly,tea polyphenols intervention and exercise intervention group of diabetic rats characterized by slightly better than the diabetic control group and intervention group exercise combined with tea polyphenols and slightly better than single intervention group,but did not reach normal rats state.2)Compared with group C,the liver index of DC rats was significantly increased(P<0.01).Compared with DC rats,the liver index of DET group was significantly decreased(P<0.05).3)Compared with the C group,a very significant increase in fasting blood glucose,insulin index of rats in the DC group(P<0.01),insulin resistance was significantly increased(P<0.05),compared with group DC,fasting blood glucose,insulin index decreased significantly in the rats of group DE resistance(P<0.05),fasting blood glucose of rats in the DT group decreased significantly(P<0.05),insulin resistance index was significantly decreased(P<0.01),were significantly decreased fasting blood glucose,insulin index of rats in the DET group(P<0.01),insulin resistance was significantly lower(P<0.05);effect of combination interventions alone than pre.4)Compared with C group,blood CRP level of rats in DC group increased significantly(P<0.05);compared with DC group,CRP level in DT group and DET group was significantly lower(P<0.05),but DE group was significantly decreased(P<0.01).5)Compared with group C,the content of MDA in the liver of DC rats was significantly increased(P<0.05),while the activity of SOD was significantly decreased(P<0.01).Compared with DC group,the DE group,DT group and DET group of rats liver MDA content were significantly decreased(P<0.05).the MDA activity in the DET group was significantly higher than that in the control group(P<0.01),and the activity of SOD in the rats of each group was significantly increased(P<0.05).Compared with DE and DT groups,the content of MDA in the liver of DET group was decreased(P>0.05),and the activity of SOD was also increased(P>0.05)6)Compared with the C group,the liver tissue of rats in the homogenate IL-6 levels were increased,while the DC group increased significantly(P<0.05);compared with DC group,DE group and DT IL-6 levels decreased significantly(P<0.05),and DET group was significantly decreased(P<0.01).7)the relative expression of IL-6 mRNA in rats was significantly higher in group DC than that of other groups(P<0.01).Compared with the C group,the relative expression of IL-6 mRNA in rats increased significantly in DC group(P<0.05);compared with DC group,DE group,DT group rats IL-6 mRNA levels were significantly decreased(P<0.05),the relative expression of IL-6 mRNA in rats in DET group decreased most obviously(P<0.01);compared with DE group,rats in the DT group,the IL-6 level of DET rats decreased(P>0.05).8)the relative expression of IL-1β mRNA in rats was significantly higher in group DC than that of other groups.Compared with C group,the level of IL-1βmRNA in DC group was significantly higher than C group(P<0.01);compared with group DC,DE and DT group in liver of rats,the relative expression of IL-1βmRNA were significantly lower(P<0.05),the relative expression of IL-1βmRNA in rats in DET group were significantly decreased(P<0.01);compared with DE,DT group,in DET group the level IL-1βmRNA has the downward trend(P>0.05).Conclusion:1)aerobic exercise and tea polyphenols alone intervention can reduce blood glucose and insulin levels in diabetic rats,both combined intervention effect is better.2)aerobic exercise and tea polyphenols alone intervention can improve antioxidant enzyme activity and improve liver oxidative damage in type 2 diabetes,the combination of both intervention effect is better.3)aerobic exercise and tea polyphenols alone intervention can reduce inflammatory cytokines of liver and decrease the liver inflammation response of type 2 diabetes,the joint effect is better. |
