Expression Of MiR-21 And TGF-β1 In The Lung Fibrosis Tissue Of ARDS Rats Induced By Sepsis

Purpose: To observe the expression of miR-21 and TGF-β1 in the lung fibrosis tissue of ARDS rats induced by sepsis and explore the role of miR-21 and TGF-β1 in sepsis-induced ARDS pulmonary fibrosis rats.Methods: 72 SD rats were randomly divided into control group(sham operation group,36 rats)and model group(experimental group,36 rats).After operation,groups were observed,such as activity,diet and respiration.ARDS was estimaed by the activity of the rats and the blood gas analysis.In experimental group,every 6 rats were sacrificed at 12 h,1d,2d,3d,4d,5d after operation,and so were the control.The left lung tissue of each rat was collected and observed by a microscope.The expression of miR-21 in lung tissues was measured through fluorescence real-time quantitative PCR.The expression of transforming growth factor-β1 were examined by immunohistochemistry staining.Then,the results were analyzed by software SPSS 18.0.Results: Compared with the control group,the activities of the experimental group were significantly decreased.The inflammation and exudation appeared in the experimental groups and pulmonary fibrosis gradually developed.The expression levels of the miR-21 from 1d of the experimental groups were higher than those in the control groups at the same time point(2d:t=7.121,P=0.002;3d:t=-5.330,P=0.006;4d:t=-6.513,P=0.003;5d:t=-19.371,P=0.000).The expression of TGF-β1 protein in the experimental group was significantly higher than that in the control group at the same time point(P<0.05).Conclusion: The expression of mi R-21 and TGF-β1 protein were increased in the lung fibrosis tissues induced by ARDS.MiR-21 and TGF-β1 may probably play an important role in the pulmonary fibrosis in ARDS rats induced by sepsis.
