Part one The characteristic of HCV infectionbetween injection drug users and general population in ChenzouObjective:To analyze the prevalence and characteristics of Hepatitis C virus(HCV)infection between intravenous drug users(IDUs)and general population.Methods: Venous blood from 1040 intravenous drug users in drug rehabilitation center and 780 cases of the general population in physical examination were collected in Chenzhou,Hunan Province,China.ELISA was uesed to detected the serum of HBV,HCV,HDV,HIV and TP infection markers.RNA of IgG positive samples was extracted by RNA extraction kit and then reverse transcripted to cDNA by reverse transcription kit.PCR amplification,sequencing and Real-time quantitative PCR were used to detecte virus RNA loading.Results: HCV infection and HCV RNA positive rate in intravenous drug users were significantly higher than that in the general population.The age distribution ofintravenous drug userswere lower than general population and male rate were higher than general population.Those who are under 40,from 41 to 50 and over the 50 years old in intravenous drug users were mainly infected by HCV 6a genotype.And in the general population those who are under 40 years old and between 41 to 50 years old were primarily infected by 6a and 3a,3b respectively,however,those who are over 50 years old were mainly infected by type 1 b.Viral loading in IDUs included various HCV genotype peoples(1 b,3 a,3 b,6 a,other)was lower than that in the general population.Conclusions:The HCV infection rate,RNA detection rate and viral load of the intravenous drug users are remarkably higher than that of the general population.Distributions of age,gender and main genotype of HCV in 2 groups had differences.Part twoAnalysis activity of HCV envelope protein binding antibody and HCV pseudo-particle neutralizing antibody in patients with hepatitis c infectionObjective: To analyze the binding activity and neutralizing activity of envelope proteins in patients with chronic HCV infection and HCV infection recovery.Methods: ELISA was used to detect antibody binding force of the serum to HCV envelope proteins of 8 genotypes,Cross neutralization ability was tested through antigen-antibody reaction with 8 genotypes of HCVpp.Results: We found that not only the relative binding and neutralization activity but also the binding width of the serum of chronic HCV infection group with HCV envelope protein E1E2 were significantly higher than tha of HCV recovery group.But there was no remarkable difference of neutralization width between chronic HCV infection group and HCV recovery group.The antibody in serum from who were infected by HCV1 b,2a,3a,3b and 6a binded with 3b envelope protein weakly.And the relative binding activity of antiboy infected by HCV 2a with 8 kind of genotypes envelope proteins was the weakest.Its binding width with antigens was also the narrowest one.However,the relative binding activity of antibody infected by HCV 1b,2a and 4a with different genotype antigen had no significant difference.Comparing the neutralization activity of serum from 1b,2a,3a,3b and 6a infected patients,3b infected group was the strongest and broadest neutralization activity,and the 2a,3a infected group were lower than other groups.The narrowest one was 2a infected group.Camparing the relative binding rate between HCV isotypes and heterogenic infected groups,all the isotypic infection was higher than the hetergenic infection group,except for 2a infection group.And the neutralization activity of all the isotypic pseudovirus with hetergenic pseudovirus was almost same.The neutralization acitivity of serum from those who had stronger HCV envelope protein binding activity was higher,same with its binding width and neutralizing wide spectrum with envelope proteins.Conclusions : The serum antibody from chronic HCV infected group had stronger and broader binding activity with HCV pseudovirus(HCVpp)than that from HCV recovery group.The HCV envelope protein binding activity and HCVpp neutralization ability of HCV infection group had remarkable genotypic difference.The neutralization ability and wide range of patients’ surum was related with its bonding strength and binding width with HCV envelope proteins.
