B7-H4 is found in 2003 by Sica.B7-H4 mRNA is widely expressed in human peripheral tissues,and B7-H4 protein,expressed in various tumor tissues,including lung cancer,ovarian cancer,gastric cancer,is considered as a tumor marker and is closely related with tumor development.In a study,immunohistochemical staining technique,Western blotting technique,and high-density tumor tissue microarray were used to detect the expression levels of B7-H4 in different tumor tissues,and high expression of B7-H4 molecules is detected in non small cell lung cancer,renal cell carcinoma,breast cancer,ovarian cancer,endometrial cancer and other malignant tumor tissues.By doing correlation analysis with clinical data,B7-H4 expression level is closely related with the pathological grading,the clinical stage,the size of the tumor in these patients.At present,the B7-H4 receptor has not been cloned,B7-H4 receptor antibody still needs a further research.B7-H4-Fc fusion protein which binds to the B7-H4 receptor,function as B7-H4 molecules.We have found that B7-H4 receptor massively expressed in tonsil tissues and lung cancer tissues tested by using the B7-H4-Fc fusion protein in flow cytometry.In the experiment,the expression levels of B7-H4 are detected in cancer tissues and paracancerous tissues of patients with pulmonary adenocarcinomas by immunohistochemical staining technique,and were analyzed with clinical pathological parameters.Part Ⅰ:B7-H4-mFc fusion protein was used to detect the expression of B7-H4 receptor in tonsil tissues,lung cancer tissues and adjacent tissues by immunohistochemical.Objective: To detect the expression of B7-H4 receptor in paraffin sections of tonsil,lung cancer and adjacent tissues,to determine the optimal concentration of B7-H4-mFc fusion protein,and to prepare for the following lung adenocarcinoma tissue microarray experiment.Methods: Paraffin sections were prepared from tonsil tissues,lung cancer tissues and adjacent tissues,and the expression of B7-H4 receptor was detected by immunohistochemistry staining technique.Results: Different degrees of brown showed in tonsil tissue,lung cancer tissue and paracancerous tissue.The staining result is best when B7-H4-mFc fusion protein concentration is 2.5ug/ml.Nucleus staining showed clear Pale brown and hematoxylin staining showed bright royal purple with staining state clearly,Clean background,no nonspecific staining.Conclusion: B7-H4 receptors were expressed in different degrees in tonsil,lung cancer and adjacent tissues,and mainly located in the nucleus.Part Ⅱ: Detection of B7-H4 receptor expression in lung adenocarcinoma tissue microarray by B7-H4-mFc fusion protein and its correlation with clinical parameters。Objective: Detection of B7-H4 receptor expression in cancer tissues and adjacent tissues of patients with lung adenocarcinoma by B7-H4-mFc fusion protein and its correlation with clinical parameters。Methods: By immunohistochemistry,the expression of B7-H4 receptor in cancer tissues and adjacent tissues of 75 cases of patients with lung adenocarcinoma is detected by B7-H4-mFc fusion protein.Scored by the staining intensity and positive rate of cells,and to analyze its correlation with clinical and pathological data of patients.Analying the result of statistics by SPSS software.Results: The expression of B7-H4 receptor in 75 cases of patients with lung adenocarcinoma was divided into different grades according to the staining intensity and positive rate.The expression of B7-H4 receptor in lung adenocarcinoma tissues was higher than that in adjacent tissues;There was no significant difference in the expression of B7-H4 receptor in adjacent tissues,and there was no significant correlation with prognosis;The correlation analysis of B7-H4 receptor expression in lung adenocarcinoma and the survival rate of patients showed that the expression of B7-H4 receptor in lung adenocarcinoma is higher,with the lower survival rate,the worse prognosis.Tumor size,T stage,N stage and clinical stage were significantly correlated with the survival time of patients with lung adenocarcinoma,and there was no significant correlation between gender,age,pathological grade and survival of patients with lung adenocarcinoma;Tumor size,T stage,N stage and clinical stage were not independent prognostic indicators of lung adenocarcinoma.Conclusion: So B7-H4 receptor expressed in lung adenocarcinoma can be considered as a negative prognostic index of lung adenocarcinoma.We still need to expand the samples to further analyze the relationship between the expression of B7-H4 receptor and the prognosis in patients with cancer,and provide the basis for clinical application.
