| Objective:Through establishing female SD rat model of postmenopausal osteoporosis by removing their ovaries,the experimental groups were given distilled water,estradiol valerate and different doses of cucumber seeds by means of intragastric administration.The purpose is to observe effect of the cucumber seeds in ovariectomized rats on osteoprotegrin,receptor activator of nuclear factor kappa β and receptor activator of nuclear factor kappa β ligand,to observe the effect of the cucumber seeds on rat body weight,hair growth.The aim of this experiment is to explain the possible mechanism of cucumber seeds on bone metabolism,and to provide theoretical and experimental evidence for the prevention of PMOP.Method:Simulation of rats model for postmenopausal osteoporosis:54 SD and SPF female rats of 7 months old were randomly divided into 6 groups according to weight: control group(Sham)(9 rats),model group(OVX)(9),positive group(estradiol valerate)(9),cucumber seeds with low dose group(CS1)(9),the dose of cucumber seeds in the treatment group(CS2)(9)and high dose treatment group(CS3)(9).After 2 weeks of adaptive feeding,the OVX group,the positive group,the CS1 group,the CS2 group and the CS3 group removed the bilateral ovarian tissue,and the Sham group excised the equal volume of abdominal fat.Drug gavage dose for 2 weeks after operation: Sham group and OVX group were fed with distilled water 0.1ml/kg·d,positive group Estradiol Valerate Tablets 0.1mg/kg·d,CS1 group 1.05g/kg·d group CS2 of cucumber seed powder,cucumber seed powder 2.1g/kg·d,CS3 group of cucumber seed powder 4.2g/kg·d,and daily records of weight and diet,the amount of drinking water in rats.After 16 weeks,the hair and body weight of the rat were observed.The maximum load of the left femur and the left tibia,the strength of the fourth lumbar and fifth lumbar vertebrae were measured by the small animal bone densitometer.The blood estrogens,OPG,RANKL and RANK were measured by the abdominal aorta,and the changes of the intrauterine membrane were observed by the ELISA staining,and the RT-PCR method was used to measure the expression level of OPG and RANK m RNA.Results:After 16 weeks of administration,compared with OVX group,rats in CS1,CS2 and CS3 group had dense,shiny,white and soft texture.The body weight of OVX group was more obvious than that in Sham group,and the body weight of CS1,CS2 and CS3 group was more stable than that of OVX group.Bone biomechanical index: compared with Sham group,the maximum load of left femur,fourth lumbar structure strength and fifth lumbar structure were significantly lower in OVX group(P<0.05),which indirectly indicated the success of postmenopausal osteoporosis model.In the left femur maximum load test group,compared with CS1,CS2 group and the OVX group,the difference was statistically significant(P< 0.001,P<0.05).Compared with OVX group,low,medium and high dose of cucumber seeds could enhance the structural strength of the fourth lumbar vertebrae In the fourth lumbar structure strength test group,the difference was statistically significant(P<0.001,P<0.001,P< 0.001).In the fifth waist,compared with the OVX group,the low,middle and high dose of cucumber seeds could enhance the strength of the fifth lumbar vertebrae,and the difference was statistically significant(P<0.001).After 16 weeks of gavage,there was no negative change in the uterus of each group.by HE staining Serological indicators: compared with the OVX group,the serum estrogen levels in the CS1,CS2 and CS3 group increased significantly(P<0.001).Compared with the OVX group,the serum OPG level of the CS1 group and CS3 group increased,and the difference was statistically significant(P<0.05).Compared with group X,the level of serum RANK expression in group CS1 and CS3 rats increased,and the difference was statistically significant(P<0.05).Compared with OVX group,the level of RANKL expression in serum was decreased in CS1 group and CS2 group,and the difference was statistically significant(P<0.05).RT-PCR measurement index: compared with OVX group,the expression of OPG m RNA was significantly higher in group CS1,CS2 group and CS3 group,and the difference was statistically significant(P<0.05),and the expression of CS3 group was significantly lower than that in OVX group.Conclusion:1,Low medium and high doses of cucumber seeds can promote hair growth in th e back of ovariectomized osteoporotic rats.2,The low,middle and high dose of cucumb er seeds can reduced the weight gain of ovariectomized osteoporotic rats.3.Gavage of the low,medium and high doses of cucumber seeds for 16 weeks had no negativeeffe ct on the uterus of ovariectomized osteoporotic rats.4,The low,middle and high dose of cucumber seeds can significantly increase the left femur,left tibia,fourth lumbar vert ebrae and fifth lumbar vertebrae in castrated osteoporosis rats.5,The cucumber seeds h ave a preventive effect on PMOP,and its preventive effect may slow down postmenop ausal bone loss by regulating OPG/RANK/RANKL system. |
