| Background:The microbial contamination has been widely concerned in recent years in Dental chair waterlines(DUWLs),once the water system is contaminated,the suspected pathogens can enter the patient’s mouth in different ways,or become water mist dissemining though the air between the departments,which is one serious potential danger to the medical personnel or the patients.The result of early routine researchs show that the dental unit water lines usually contain staphylococcus streptococcus and actinomycetes and the common E.Coli.According to current practical standard,the DUWLs in every single hospital anti-contamination is far from the requirements for aseptic in microbiology index.And in the use of each dental chair has its’ own disinfection mechanism and every single one’s own different frequency and years of use.So,how to prevent the contamination of bacteria from DUWLs and what kind of disease should we control,these are the mainly problem that needed to in-depth study.So,the serious pollutions in DUWLs are no longer just the partly drilling disinfection problems,but it become the important risky factor for iatrogenic infection.Objective:The purpose of this study is to investigate the pollution of oral bacterias in the high-speed hand pieces and its’ waterlines system in one big Stomatology hospital dental chair.Take this to understand the status of the hand pieces’ waterline contamination with bacteria,even with particularity bacteria,to explore the necessity of further improvement of disinfection methods and relevant strategies.Methods:Sample 20 High-speed air turbine hand piece water unit from dental chair of water outlet,after routine disinfection of dental treatment chair.And also 1 sample from distilled water from the laboratory.Then seal them in-4℃ environment for the experiments.5 randomly selected samples for applying Illumina MiSeq pyrosequencing for analysing random drawing for the bacteria in the first part of this study.After the 16s sequencing,we tested the volume of Burkholderium and Proteobacterium in dental unit water lines by polymerase chain reaction(PCR).Results:Through the general culture method,the results show that in the "10" second group has the most bacterium which are also under the requirements of the national standard And the result of 16sDNA shows that the dental unit water lines contain staphylococcus,streptococcus,actinomycetes and the common E.Coli.The PCR experimental results showed Burkholderia and Proteobacterium in dental unit water supply system was significantly increased,these two kinds of bacteria and the higher frequency of dental ch air uses have positive correlation.Among 20 samples,the ultrasonic scaler has the most higher detection than the other dental chair.While the negative control results(distilled water)found that in Burkholderia or Proteobacterium does contain a phase of high concentration in dental unit water lines.The comparison between Burkholderia and Proteobacterium is statistical significance.(P<0.05)Conclusion:The waterway system in dental chair does exist Burkholderia and Proteobacterium contamination.Those two bacterium’s volume may relate to the frequency of water use.How to prevent bacterial contamination and the main kind preventing of bacteria and treatment of the disease should be further studied and discussed.In summary,it has been proved that the serious pollution of dental water system,is not just part of the drill disinfection problem,but the important most factor of nosocomial infection.The disinfection of the dental use of water and the prevention and control is imminent.And in the results of PCR,we have learnd that the content of these particularly bacterium are much more in scaling than other sample sources.And compared to the blank control,these two bacterium have marked increase.So,Burkholderia and Proteobacterium have the high volume in dental treatment chair in the water. |
