Effect Of MicroRNA-125a-5p Expression On Clinical Prognosis And Activation Of NF-kappa B Pathway In Diffuse Large B Cell Lymphoma

Objective: To detect the expression of miR-125a-5p in diffuse large B cell lymphoma(DLBCL),investigate the relationship between the expression of miR-125a-5p and Nuclear factor kappa B/P65(NF-κB/p65),apoptosis related proteins,as well as the clinicopathological features and prognosis of DLBCL.Methods: A total of 84 cases of DLBCL,who were diagnosed in the department of pathology of the affiliated hospital of Guizhou medical university,were collected.At the same time,10 cases with reactive hyperplasia of lymph node were used as the control for detecting mi R-125a-5p.The expression level of miR-125a-5p was examined by SYBR Green real-timereverse transcriptase polymerase chain reaction(real-time RT-PCR).The expression of NF-κB/p65,Bcl2,Caspase3,Ki-67 were detected by immunohistochemical stain.Collection of the clinicopathological data and follow-up were done.SPSS19.0 was used to statistically analyze Results: The expression of miR-125a-5p in the DLBCL was up-regulated,which was 4.99 ± 8.22 folds in reactive hyperplasia samples(P= 0.002).The expression of mi R-125a-5p in the non-germinal center B cell like-diffuse large B cell lymphoma(non-GCB DLBCL)was significantly higher than that in germinal center B cell like diffuse large B cell lymphoma(GCB-DLBCL),P= 0.027.Comparing with reactive hyperplasia of lymph node,the expression of NF-κB/p65 [10%(1/10);58%(49/84)],Bcl2 [30%(3/10): 68%(57/84)] in DLBCL were obviously increased.Whereas exression of caspase3 reducesd in DLBCL.P-value was respectively 0.004,0.019,0.038.Expression of NF-κB/p65,Bcl2 and Caspase3 protein(42/65,48/65,32/65)were higher in high expression of miR-125a-5p than low miR-125a-5p in DLBCL and p-value was respectively 0.031 、 0.030 、 0.022;there was no significant difference in the proliferation of tumor cell between high and low expression of miR-125a-5p(P=0.56).Univariate survival analysis showed that high expression of miR-125a-5p was related to the prognosis of DLBCL patients(P=0.038).Multivariates regression analysis indicated high expression of miR-125a-5p might be an independent risk factor of DLBCL patients’ prognosis(P=0.013).Conclusion: The expression of miR-125a-5p in DLBCL is up-regulated,and the expression in non-DLBCL is more obvious.It is an independent risk factor for prognosis in DLBCL patients.The upregulation of miR-125a-5p expression is related to the activation of NF-kappa B,the expression of Bcl2 and Caspase3 proteins in DLBCL,and the activation of NF-kappa B by miR-125a-5p may be a molecular mechanism by which Bcl2 expression increases and Caspase3 expression decreases.
