| Staphylococcus aureus(S.aureus)is a kind of important gram-positive pathogens that cause human infection and food poisoning.The clinical harm to S.aureus is serious.S.aureus infection is especially important to the elderly,postoperative patients,patients with weak diseases,and newborns.It is easy for that antibiotic to produce multiple drugs resistance,which causes people to choose the vaccine immunization vaccination to prevent s.aureus bacteria infection.There have been studying that have shown that Target of RNAIII Activating Protein(Trap)is highly conservative in S.aureus,and has a certain immunogenicity and immune protection.While Trap has become an important candidate protein for the development of s.aureus vaccine,the immunological effect remains unsatisfactory.Therefore,it has important scie ntific significance and application value to explore how to improve the vaccine efficacy of S.aureus.As a safe,efficient multifunctional herbaceous plant with strong immunoregulation activity,garlic has attracted much attention in recent years.However,after chewed and broken,alliin and allinase contact each other,to generate the allicin has strong garlic smell and after eating bad breath left for a long time,are unacceptable.Compared with garlic,black garlic has a good taste,no garlic odor and a slightly sweet taste,which can be eaten directly without extra processing.It was found that,compared with common garlic,black garlic has a higher antioxidant activity,and it has an important role in anti-sporty fatigue,inhibiting the growth of cancer cells and metastasizing,anti-inflammatories,and improving the ability to learn.It’s also noted that the black garlic has an obvious immunomodulatory function.So far,the research about black garlic immune response mainly concentrated on the nonspecific immune response and obtained the certain research results.However,there is little research on the specific immune response to black garlic,and the effect of black garlic on immune response after vaccination has not yet been reported.In this study,BALB/c mice were studied to investigate the effects of black garlic on the immune response to S.aureus Trap vaccine after immunization,aiming to explore the immune response that promoted the stronger and broader induction of S.aureus vaccine.180 female mice were randomly assigned to adjuvant control group,Trap control group,low and high dosageblack garlic treatment group and low and high raw garlic treatment group.Four treatment groups were given different dosages of black garlic or raw garlic extract for 28 days,adjuvant control group and Trap control group was received equivalent saline solution,and treatment groups were vaccinated twice with S.aureus Trap in two-week intervals.Then,the lymphocyte was collected from spleen of Trap immunized BALB/c mice and the ability to induce T-cell proliferation and cytokines response were detected.Two weeks after boosting,blood samples were collected,then trap specific IgG levels and IgG subclass reactions were measured.The results showed that high and low black garlic extract significantly enhanced the serum Trap-specific IgG and IgG2 a level,up-regulated the secretion of IFN-γ and IL-17 a,improved the immune protective effect.It was suggested that the extract from black garlic ethanol extract could promote staphylococcus aureus Trap protein induced stronger cellular immune response and humoral immune response and induced the better resistance to staphylococcus aureus Newman strains infection.This research provides theoretical basis for the practical application of black garlic and provides experimental basis for the application of black garlic to improve the immune effect of other vaccines. |
