| Objective: The aim of this study was to investigate the expression level of CUL4 A in non-small cell lung cancer(NSCLC)tissues and paracancerous tissues by immunohistochemical method,and to explore its relationship with clinicopathologic characteristics and clinical outcomes of NSCLC.Methods: 120 cases of NSCLC tissues were screened from October2012 to December 2014,and 30 cases of adjacent normal tissues were selected as control.All patients with data and clinical and pathological information are complete.All of the above pathology diagnosis by our hospital two pathologists identified.All patients underwent radical surgery,and all patients with stage Ⅲ A had no neoadjuvant chemotherapy before surgery.The expression of CUL4 A in NSCLC tissues and adjacent tissues was detected by immunohistochemistry(SP method).To analyze the correlation between CUL4 A expression and clinicopathological data(including sex,age,smoking,pathology,clinical stage,histological grade,lymph node metastasis)and prognosis.Result:(1)The positive expression rate of CUL4 A in NSCLC tissues was 67.5%(81/120),which was significantly higher than that inadjacent normal tissues(26.7%,8/30).There was significant difference between the two groups(p=0.007).(2)There was no significant correlation between the expression of CUL4 A and the age and histological grade of the patients(p<0.05),but there was no significant correlation with the sex,smoking status,pathological type,clinical stage and lymph node metastasis(p>0.05).(3)The expression of CUL4 A was correlated with the median OS in NSCLC patients.The median OS of CUL4 A positive expression group was 23.9 months,and the median OS was not reached(p=0.003),in negative expression group,but had no significant correlation with median PFS(p>0.05).The expression of CUL4 A was associated with median OS in patients with lung squamous cell carcinoma.The median OS of CUL4 A positive group and negative expression group was 23.8 months and 28.2 months(p<0.01),and there was no significant correlation with PFS(p>0.05).There was no significant correlation between the median PFS and the median OS in patients with lung adenocarcinoma(p>0.05).The expression of CUL4 A was correlated with the median OS in patients with stage Ⅰ and Ⅱ stage.The CUL4 A positive expression group and the negative expression group(p=0.008),and there was no significant correlation with the median PFS(p>0.05).The expression of CUL4 A was not significantly correlated with the median PFS and median OS in patients with stage ⅢA(p>0.05).The expression of CUL4 A was differentiated from grade Ⅱ and grade Ⅲ(p<0.05).The median PFS of CUL4 A positive expression group was 16.4months,the median PFS of negative expression group was not reached(p=0.044),the median OS of CUL4 A positive expression group was 24.3months(p<0.01).There was no significant correlation between CUL4 A expression and median PFS and median OS in patients with grade I differentiation(p>0.05).Conclusion :(1)The expression of CUL4 A in NSCLC was significantly higher.(2)The expression level of CUL4 A was correlated with age,histological grade and prognosis of patients,but not with gender,smoking status,pathologic type,clinical stage and lymph node metastasis,and could be used as a prognostic predictor of NSCLC reference one of the indicators. |
