| Objective:Study on gastroparesis kangfang and domperidone treatment of diabetic gastroparesis rats,by observing the general condition of rats,body weight,blood glucose,gastrointestinal emptying rate and motilin and gastrin levels recorded,to explore the influence of gastroparesis Kang on diabetic gastroparesis rats and its possible mechanism,and provide experimental basis for clinical treatment of traditional Chinese medicine DGP.Methods: Healthy SD rats,male,60,weighting 200-220 g,fed with common feed for 1 weeks.1 weeks later,the rats were randomly divided into normal control group(n = 12),and the other 48 rats were used to replicate the DGP model.Modeling by STZ modeling method,the rats were randomly divided into four groups: model group,domperidone group,gastroparesis kangfang low dose group,gastroparesis Kang side high dose group,12 rats in each group.The normal control group were fed with normal diet rules,and in the model group,western medicine group were given high fat diet(normal diet plus lard mixture)of feeding,that day morning meal,afternoon eating shuangri.The rats in each group were free to drink water.Each group was orally administered after the success in the modeling,domperidone group,gastroparesis kangfang low dose group and gastroparesis Kang Fanggao dose groups were given domperidone suspension,low dose and high dose of gastroparesis Kang liquid gastroparesis Kang liquid gavage,the normal control group and model group were given equal volume of distilled water the stomach,all the rats by gavage for 6weeks.During the experiment,the general condition of the rats was observed and recorded.After the last injection,the rats were fasted for 24 hours,giving the phenol red solution by gavage,intraperitoneal injection of chloral hydrate anesthesia,abdominal aortic blood,on the determination of motilin and gastrin levels in rats and hormone radioimmunoassay;laparotomy was performed to remove the entire stomach,the stomach contents,to determine the gastric emptying rate.Results:1 The general condition of experimental rats The rats in the normal group had normal activity,quick response,no significant changes in the amount of food intake,water intake,urine output,stool traits,and so on.Model group,domperidone group,gastroparesis kangfang low dose group and high dose group of gastroparesis kangfang rats appeared yellow fur rough,dull,lack of energy,food intake,water intake,urine and stool quantity gradually reduce the phenomenon after more than before,but the model group is more obvious,in general Martin oxazoline group,gastroparesis kangfang low dose group,high dose group kangfang gastroparesis rat has improved compared with the model group.2 The weight change of rats Model group,domperidone group,gastroparesis kangfang low dose group and gastroparesis kangfang weight of rats in high dose group decreased significantly compared with the normal group,the difference was statistically significant(P < 0.05);compared with the model group,domperidone group,gastroparesis kangfang low dose group and gastroparesis weight kangfang rats in high dose group increased,the difference was statistically significant(P<0.05);Compared with the domperidone group,gastroparesis kangfang low dose group and high dose group of gastroparesis kangfang weight increased,the difference was statistically significant(P<0.05);gastroparesis kangfang in low dose group and high dose group compared with each other,the difference was not statistically significant(P>0.05).3 The changes of glucose in rats Compared with the normal group,model group,domperidone group,gastroparesis kangfang low dose group and high dose group kangfang gastroparesis rats blood glucose levels were significantly increased,the difference was statistically significant(P<0.05);compared with the model group,domperidone group,gastroparesis kangfang low dose group and the blood glucose level of gastroparesis Kang Fanggao dose group rats decreased slightly,but the difference was not statistically significant(P > 0.05);gastroparesis kangfang domperidone group,low dose group and high dose group of gastroparesis kangfang 22 comparison,no statistically significant difference(P > 0.05).4 The rate of gastric emptying in rats Model group,domperidone group,gastroparesis kangfang low dose group and high dose group kangfang gastroparesis rat gastric emptying rate decreased significantly compared with the normal group,the difference was statistically significant(P<0.05);compared with the model group,domperidone group,gastroparesis kangfang low dose group and high dose of gastroparesis Kang rats,the gastric emptying rate was significantly increased,the difference was statistically significant(P<0.05);compared with the domperidone group,gastroparesis kangfang low dose group and high dose group kangfang gastroparesis gastric emptying rate increased,the difference was statistically significant(P < 0.05),gastroparesis Kang side high dose group was higher than that of the stomach Tan Kang side low dose group,the difference was statistically significant(P < 0.05).5 The expression level of MTL in rats Compared with the model group,domperidone group,gastroparesis kangfang low dose group and high dose group of gastroparesis kangfang MTL content decreased obviously,the difference was statistically significant(P<0.05),gastroparesis kangfang low dose group and high dose group of gastroparesis kangfang MTL were lower than domperidone group,the difference was statistically significant(P<0.05),gastroparesis kangfang low dose group and high dose group between the two groups had no significant difference(P<0.05),compared with the normal group,no significant difference(P<0.05).6 The expression level of GAS in rats Compared with the model group,domperidone group,gastroparesis kangfang low dose group and high dose group of gastroparesis kangfang GAS was significantly decreased(P< 0.05);compared with the domperidone group,gastroparesis kangfang low dose group and high dose group of gastroparesis kangfang GAS content decreased,the difference was statistically significant(P < 0.05),gastroparesis kangfang low dose group and high dose group of gastroparesis kangfang two there was no significant difference between groups(P > 0.05),the two groups compared with the normal group,the difference was not statistically significant(P > 0.05).Conclusion:1 The diabetic gastroparesis rat model was successfully replicated,and through the determination of gastric emptying rate,it was verified that diabetic gastroparesis rats do exist gastrointestinal motility disorder.2 Gastroparesis Kangfang can improve the rate of gastric emptying in diabetic gastroparesis rats,and the effect was better than domperidone.3 The treatment of DGP may improve the level of MTL,GAS and other gastrointestinal hormones in DGP rats,which provides a scientific basis for the treatment of DGP. |
