| Objectives:This study aims to investigate the expression of Wnt/β-catenin signaling pathway in the pathogenesis and progression of pulmonary fibrosis by detecting the expression of β-catenin and Gsk-3β in normal lungtissue and fibrotic lung tissue,an d the effect of hydrogen sulfide Sulfide,H2S).Methods:Sixty healthy male SD rats were randomly divided into four groups: control group(group A),BLM group(group B),BLM + Na HS group(group C),prednisolone group(group D).B、C、D group were perfused BLM solution(5mg/kg)to establish p ulmonary fibrosis model,group A was injected with equal volume of saline.The rats in group C were injected intraperitoneally with NaHS solution(28μmol/kg),group D was injected with prednisolone solution(0.56mg/kg)and group A and group B were injected with equal volume of normal saline.Five rats were sacrificed at the 7th,14 th and 28 th day after modeling.HE and Masson were stained with 4% formaldehyde in the left lungtissue of rats.The degree of alveolitis and pulmonary fibrosis was observed and detected by immunohistochemistry The expression of β-catenin and Gsk-3β protein in the right lung tissue was detecte d by RT-PCR.Results:1.HE and Masson staining showed that the lung tissue fibrosis wasthe lowest in the control group and the BLM group was the most important.The lung tissue of BLM+NaHS group and BLM+prednisolone group also had alveolitis and fibrosis,but the degree Were significantly less than BLM group.2.The results of immunohistochemistry showed that β-catenin and Gsk-3β protein were low in the lung tissue of the control group.BLM group was significantly higher than that of control group(P<0.05).The expression of β-catenin and Gsk-3β in BLM+Na HS group and BLM+prednisolone group were higher than those in control group,which was significantly lower than that in BLM group.The difference was statistically significant(P<0.05).3.The results of RT-PCR showed that the expression of β-catenin and Gsk-3βm RNA was the lowest in the lung tissue of the control group.BLM group compared with the control group was significantly increased,the difference was statistically significant(P<0.05).The expression of β-catenin and Gsk-3β in BLM+Na HS group and BLM+prednisolone group was higher than thatin control group at each time group,which was significantly lower than that in BLM group(P<0.05).Conclussions: Exogenous H2S can inhibit the expression of β-catenin and Gsk-3β in lung tissue of rats,so it can inhibit bleomycin-induced pulmonary fibrosis in rats.The mechanism may be related to the activation of Wnt/β-catenin signaling pathway. |
