Background: Mitotic arrest deficient 2(MAD2)controlling the function of spindle assemble checkpoint(SAC),is expressed in various kinds of cancer,yet the expression of MAD2 in small cell lung cancer(SCLC)maintains vague.Anti-microtuble spindle antibody(MSA)and anti-centromere antibody(ACA)are two major types of autoantibodies,whose targeted antigens act on vital roles in mitosis.Current researches has provided evidences that ACA was detected in SCLC.However,the expression of MSA and ACA are not clear in SCLC.Objective: The object combining MAD2 with MSA and ACA was to explore their clinical value for the diagnosis of SCLC,providing clinical evidences for molecular studies of SCLC.Methods: 120 SCLC patients,110 non-small cell lung cancer(NSCLC)patients,115 Pulmonary Nodule(PN)patients and 110 health group(HG)patients were enrolled in this study.And MAD2 expression,was tested by qt-PCR.MSA,ACA were detected by indirect immunofluorescence(IIF).Results: MAD2 was overexpressed in SCLC and NSCLC(P<0.05).Interestingly,there were significant differences between SCLC and NSCLC using qt-PCR(P<0.05).AUC under ROC curve of MAD2 expression was 0.799 with medium diagnostic value.In correlative analysis,Odds Ratio of MSA and ACA was respectively 6.94 and 5.60.In the correlation analysis,Kappa of MAD2 with MSA was 0.49,and Kappa of MAD2 with ACA was 0.42.In the parallel analysis,the sensitivity was 83.31%,the specificity was 79.34%,and the Youden Index was 0.62.Moreover,in the serial analysis,the sensitivity was 65.32%,the specificity was93.35%,and the Youden Index was 0.59.Conclusion:In comparison with NSCLC and PN,MAD2 was overexpressed in SCLC.MAD2 ought to play a critical role in the process of SCLC.The combination MAD2 with MSA and ACA expression contributing to enhance the sensitivity and specificity,has some application value for early diagnosis,clinical diagnosis and promising treatments of SCLC.
