ObjectiveCaffeine is one of the important causes of low weight infants,but the specific mechanism is not completely clear.The main purpose of this study was to explore whether excessive intake of caffeine in the early pregnancy could affect the absorption of nutrients in the yolk sac in the early pregnancy by using the yolk sac model of the embryo,which will affect the growth and development of the embryo.The preliminary exploration and study of the effects of caffeine intake on pregnant women,the impact on the fetal development speed and the mechanism of their occurrence,thus providing scientific basis for the safe and rational diet of women in pregnancy,and the realization of the aim of the eugenics and the improvement of the quality of the population,are of important ethical value and present.Practical guidance.MethodsIn this study,chicken embryos were divided into normal group,low dose group,middle dose group and high-dose group.After the fresh seed eggs were grouped at 37 degrees for 2 days,the eggs were opened on the eggshell and given different doses of caffeine on the yolk membrane.The control group was given the same volume of PBS,and then sealed with breathable tape,and the embryo and yolk tissue were taken out for 6 days.According to the morphological analysis of the egg yolk tissue through HE staining,the changes of p185 and p21 during embryo development were analyzed by RT-PCR,and the cell proliferation and apoptosis of yolk membrane were further analyzed by immunofluorescence staining.Results1.Compared with the control group,the weight of embryos decreased with the increase of caffeine concentration.The difference between the normal group and the middle dose group was statistically significant(P<0.05),and there was a significant difference between the normal group and the high dose group(P<0.01).2.The results of yolk HE staining were less than those in the control group.The yolk vascular development of the middle dose group and the high dose group was poor,and the blood vessel density decreased significantly.After H.E staining,there was obvious difference in the development state of the tissue morphology in each experimental group.3.PT-PCR experimental results compared with the control group,the expression of P185 in the experimental group decreased with the increase of caffeine concentration.The p21 RT-PCR results showed that the expression of cell cycle inhibition gene in the yolk membrane increased with the increase of caffeine dose.4.The proliferation and apoptosis of yolk tissue cells were compared with that of the control group.PH3 staining showed that the cell proliferation ability of the yolk membrane decreased with the increase of exposure to caffeine,while the result of TUNEL staining was the opposite.With the increase of caffeine concentration,the degree of apoptosis increased.ConclusionsCaffeine causes vascular distribution density to decrease,vessel diameter decreases and development becomes worse by affecting the development of the vasculature of the vitelline membrane of the embryo.At the same time,the proliferation of yolk cells is weakened and the degree of cell apoptosis is increased,which affects the absorption and transport of the yolk membrane to nutrients,and thus affects the development of the embryo.In order to prove that early high-dose intake of caffeine in pregnant women can provide data support by affecting the development of placental villus and affecting the absorption of nutrients and affecting the normal development of embryos.
