Effects Of Evening Primrose Oil On P38MAPK,NF-κB Pathway And Serum IL-1α And IL-6 In Rabbit Auricular Acne Models

ObjectiveTo observe the effect of Evening primrose oil on the treatment of rabbit ear acne model and its effects on p38 MAPK,NF-κB signaling pathway and serum IL-1α and IL-6.MethodsCoal tar coating rabbit ear in the manufacture of rabbit ear acne model,1 times a day,15 days in a row,after the success of the building were randomly divided into normal group,positive control group,medication group(Evening primrose oil high,medium and low dose group,Tanshinone group).The administration group was given once a day according to the corresponding medicine and dosage,and after 15 days treatment,it was first evaluated by macroscopic observation and then treated with HE staining to determine the therapeutic effect.The levels of IL-1α and IL-6 in serum were measured by ELISA,and the expressions of NF-κB and p38 MAPK proteins were detected by immunohistochemistry.ResultsAfter the completion of modeling diagnosed as moderate(grade Ⅱ)acne.Pathological biopsy shows the epidermis,cuticle thickening with keratosis,stratum granulosum and stratum spinosum are hypertrophy.Adjacent part of the hair follicles fused with each other and expand,hair follicles keratosis,funnel Department became large pot-shaped.Dermal capillary telangiectasia,scattered inflammatory cells can be found around the hair follicles.Increased sebaceous glands,increased with epithelial hyperplasia.The levels of IL-1α and IL-6 in the blank control group and the model group were compared,the difference was statistically significant(P <0.010),model group increased;The levels of IL-1α and IL-6 in model group and medication group were compared,the difference were statistically significant(P <0.010),each medication group decreased.Comparison of average optical density of p38 MAPK and NF-κB between model group and blank group,the difference was statistically significant(P <0.010),model group increased;Comparison of average optical density of p38 MAPK and NF-κB between model group and medication group,the difference was statistically significant(P <0.010),the medication group decreased.ConclusionEvening primrose oil,curative effect is in the treatment of acne,can reduce the rabbit ear hair follicles keratin plug formation,lighten the cuticles,inhibiting sebaceous gland ductal epithelial hyperplasia,reduce inflammatory cells infiltration.The mechanism is to control the serum IL-1α and IL-6 through p38 MAPK and NF-κB signaling pathway.
