| ObjectiveTo investigate the protective effect of folic acid combined with l-carnitine on vascular endothelium in preeclampsia rats.MethodsIn November 2016 to February 2017,we purchase 50 healthy female SD rats(Sprague wrote-Dawley),10 in each group were random Ly divided into normal control group,model of preeclampsia group,L-NAME + folic acid group(3 mg/kg·d),L-NAME + l-carnitine group(400 mg/kg·d),L-NAME + folic acid+l-carnitine group(3 mg/kg·d + folic acid l-carnitine 400 mg/kg · d).Pregnancy0.5 ? 20 days,lavage for folic acid,l-carnitine,pregnant 9?20 days back neck subcutaneous injection of 75 mg/kg·d the nitro L arginine methyl ester(L-nitro-arginine methylester,L-NAME)induced model construction of rats with preeclampsia samples,determination of rats during pregnancy 5,10,14,18 days 24h urine protein,blood pressure and small animal ultrasonic testing during the 19 th day of femoral artery blood flow mediated vasodilatation function(flow-mediated dilation,FMD),cutting palace pregnancy 21 days out,abdominal aorta in blood,using flow cytometry to detect vascular endothelial microparticles level,HE dyeing observation groups of kidney and placental tissue pathological changes,the late ELISA method to detect the plasma homocysteine(Hcy)and asymmetric dimethyl arginine(ADMA)levels and oxidative stress related factorsmalondialdehyde(MDA),8-different prostaglandins(8-iso-PGF2 ?),Bio Legend LEGENDplexTMMulti-analyte flow assay kit to detection inflammation related factors: monocyte chemotactic protein 1(MCP-1),interleukin 1 beta(IL-1 beta)and interleukin 6(IL-6).ResultsStatistical analysis of repeated measurement data of the systolic blood pressure of each group showed that the interaction of grouping,time and grouping and time were statistically significant(F=59.274,273.575,14.814,all P(27)0.05).Statistical analysis of the repeated measurement data of 24 h urine protein in each group showed that the interaction of grouping,time and grouping and time was statistically significant(F=64.104,291.911,29.533,all P(27)0.05).Compared with model group,the blood pressure and 24 h urine protein at 10 th,14th and 18 th days during pregnancy were decreased in combined group[blood pressure:(125.84±3.94)vs(141.17±2.82),(123.61±3.69)vs(152.23±1.21),(116.72±4.64)vs(146.73±1.36)mm Hg;urine protein:(6.57±0.19)vs(7.79±0.46),(8.87±0.57)vs(11.84±0.76),(8.93±1.57)vs(11.92±0.83)mg/24h;all P(27)0.05].HE staining found kidney: blank group of pregnant rat renal glomerular structure clear,model group of pregnant rats appear edema glomerular enlargement,structure,the renal capsule cavity extremely narrow,are almost glomerular fill.Placenta: the placental villi of the blank group are rich in blood vessels and clear in structure.The trophoblast cells in the placental villi are dominated by a combination of trophoblast cells,and cell trophoblast cells are less.However,the number of placental villi in the model group decreased,and the structure was fuzzy and atrophic,and some of the villous cellulose samples were necrotic,and the growth of the placental villi was increased,and most of the villi were immature.Small animals ultrasonic testing in the rat femoral artery FMD results show: ischemia reperfusion after 5 min,with theincrease of perfusion time,rat femoral artery diameter increases,FMD increased gradually,and then slow down,the folic acid group,l-carnitine,two joint group rat femoral artery FMD peaked in the 120 s,and the model group in the 300 s to reach their peak.The FMD peak level was higher in folic acid group,left carnitine group and two drugs than the model group[(16.14±0.88)%、(17.61±2.38)%、(21.21±0.79)%vs(12.20±1.92)%,allP<0.05].Compared with model group,vascular endothelial microparticles levels[(4.53±0.65)% vs(25.76 ± 2.90)%,P(27)0.05],plasma Hcy levels[(5.04 ± 1.21)vs(7.88 ± 1.52)nmol/m L,P(27)0.05] and malondialdehyde levels[(6.33±0.57)vs(12.68±0.55)nmol/m L,P(27)0.05] were all decreased in combined group.ConclusionsThe combined application of folic acid and l-carnitine can protect the vascular endothelium of PE rats and prevent the occurrence of PE by reducing the plasma Hcy and improving the level of oxidative stress injury and inflammatory factors.Because the vascular endothelial lesion of pre-eclampsia precedes the clinical manifestations,the detection of vascular endothelial lesion is conducive to the early prediction of pre-eclampsia.In the middle of a pregnancy even through FMD and EMPs high-risk pregnant women in early pregnancy screening out of preeclampsia,folic acid combined l-carnitine to intervention,to reduce maternal and infant mortality in the perinatal period has a certain clinical significance. |
