Objective: To explore the ethology,the neurotransmitters and neurons in the hippocampus of rats with vascular dementia(short for VD)treated by Modified Shengjiang powder based on the theory of tonifying kidney and purgating Fuorgans in order to provide experimental data for its clinical use.Methods: According to the random number table,90 SD rats were randomly divided into 6 groups: sham operation group,model group,high dose group,middle dose group,low dose group,huperzine group.There are 15 rats in each group.Blocking the vascular dementia rat model was established by 2 vascular blocking method.After the model was completed,the rats of unsuccessful and dead were removed from the model,and 12 rats in each group were randomly selected as the research object.High,medium and low dose group were 14ml/kg,7ml/kg,3.5ml/kg in accordance with intragastric administration of 0.8g/ml flavored water decoction,huperzine group treated with huperzine suspension 0.1mg/kg,sham operation group and model group were lavaged with physiological saline 7ml/kg,tid,28 days.After treatment,Morris water maze was used to detect the behavior and morphology of hippocampus was observed under microscope by Nissl staining.The expression of SP and VIP in the hippocampus and colon of rats were testing by ELISA.The content of MDA in rat hippocampus was testing by TBA.The content of T-SOD in rat hippocampus was testing by hydroxylamine method.The expression of SP in the hippocampus of rats was testing by ELISA.The content of AchE in rat hippocampus was testing by spectrophotometer.Western blotting was used to detect the expression of Bcl-2,Bax and Caspase-3 protein in the hippocampus of rats.Result:(1)The characteristics of praxiology: Compared with the sham operation group,in model group,the time of escape latency was extended(P<0.05),the number of learning times was decreased(P<0.05),and the number of times in the target area and platform was decreased(P<0.05),the time of staying in the target area and platform was reducing(P<0.05).Compared with the model group,the high,medium,low and huperzine group had a shorting time of escape latency(P<0.05),an increasing number of learning times(P<0.05),an increasing number of times through the target area and platform(P<0.05),an extending time to stay in the target area and platform(P<0.05).(2)The morphological characteristics of neurons: The neurons of the hippocampal CA1 region in the sham operation group were basically normal.But the neurons in the hippocampal CA1 area were loosely arranged and lost significantly in the model group.Compared with the model group,in the high,medium,low and huperzine group,the morphology of hippocampal neurons in rats was improved and arranged in a more tight and orderly manner.(3)Changes in the content of oxidative damage factors: compared with sham operation group,in the model group,the content of T-SOD in hippocampus of rats was decreased(P<0.05),while the MDA increased(P<0.05).Compared with the model group,in the high,medium,low and huperzine group,the content of T-SOD in hippocampus of rats was increased(P < 0.05),while the MDA decreased(P<0.05).(4)Changes of neurotransmitters: Compared with sham operation group,in the model group,the content of SP,VIP,Ach and AchE in hippocampus of rats were decreased(P<0.05),Compared with the model group,in the high,medium,low and huperzine group,the content of SP,VIP,Ach and AchE in hippocampus of rats was increased(P<0.05).Compared with sham operation group,in the model group,the content of SP and VIP in colons of rats were decreased(P<0.05),Compared with the model group,in the high,medium and low dose group,the content of SP and VIP in colons of rats was increased(P<0.05),while huperzine group had no difference.Compared with huperzine group,in the high,medium and low dose group,the content of SP and VIP in colons of rats was increased(P<0.05).(5)The expression of protein: Compared with the sham group,the expression of Bcl-2 protein in the hippocampus of the model rats was decreased and the expression of Bax and Caspase-3 protein was increased(P<0.05).Compared with the model group,in the high,medium,low and huperzine group,the expression of Bcl-2 protein in the hippocampus was increased,whil the Bax and Caspase-3 protein was decreased(P<0.05).Conclusion: Modified Shengjiang powder can improve the cognitive impairment caused by vascular dementia.The possible mechanism might be that it can improve the cognitive function by improving the content of Ach and AchE in hippocampus and improve the content of SP and VIP in the hippocampus to improve the cognitive function by improving the brain and intestine at the same time.In addition,it can raise the Bcl-2 protein level and decrease Bax and Caspase-3 protein level in the hippocampus to improve the apoptosis of neurons in hippocampus and improve learning memory by antioxidant effect because of improving the content of T-SOD and reduce the content of MDA in the hippocampus.
