Effect Of Yinqin Qinggan Decoction On Blood Fat And Liver Tissue Pathology In Nonalcoholic Hepatitis Rats

[Research purposes]A non-alcoholic steatohepatitis model was established by induction of a high-fat diet to observe the effects of Yinqin Qinggan decoction on blood lipids,liver function and liver histopathology in non-alcoholic steatohepatitis rats.The mechanism of action of alcoholic steatohepatitis provides a new experimental basis for the clinical treatment of non-alcoholic steatohepatitis.[Research operation]Fifty SPF SD male rats were divided into five groups according to their weight according to the random number table method.They were blank control group(blank group),model group,reductive glutathione group(Western medicine group),Yinqin Qinggan.Low-dose group(low-dose group)and high-dose Yinqin Qinggan decoction(high-dose group)were used in each group.One week after adaptive feeding,the rats were modeled,the blank group was given normal feed,and the other groups were fed high-fat diet to prepare the NASH model.After the model was completed,except for the blank group and the model group,the rats were given normal saline perfusion,and the low-dose group was given Yinqin Qinggan Decoction 0.5g(crude drug)/ml gavage,and the high-dose group was given Yinqin Qinggan Decoction.Concentrate 1 g(crude drug)/ml was given to the stomach,and western medicine group was given 0.02 g/ml of reduced glutathione solution.The experiment was carried out until the 4th weekend.Five rats were randomly selected from each group and fasted for 24 hours after intragastric administration.After weighing,chloral hydrate was used for intraperitoneal injection(rat weight per 100 g,chloral hydrate 0.3 ml).After the onset of anesthesia,take the blood from the eyeball rapidly,and place the whole blood sample at room temperature for 2 hours and centrifuge.The rotation speed is 1000 revolutions per minute.Then take the supernatant and store it in the refrigerator at-80℃.Make the date mark ready for testing.Testing of Serum triglyceride(TG),cholesterol(TC),aspartate aminotransferase(AST),and alanine aminotransferase(ALT)levels.After the blood was taken,the liver was quickly removed,the morphology was observed,and the liver wet weight was weighed to calculate the liver index(hepatic wet weight/total weight × 100%);then,the size of the right lobe tissue was taken as 0.5 cm×0.5 cm×0.5 cm.Put 10% formaldehyde solution to fix the specimen and mark the date.Put it in the refrigerator and wait for the test.On the 8th week of the experiment,the remaining rats were intragastricly fasted for 24 hours and the specimens were collected in the same manner as described above.The samples taken above were sent for testing for statistical analysis.[Research result](1)The model group was compared with the blank group: the weight of the rats increased,the hair was rough,the body size was significantly increased,and the spirit was apathetic,and the daily activities were reduced.Visually observing the dark yellow color of the liver tissue of rats,increasing the volume,and adhesion with peripheral organs,there is no obvious edge line;liver index,serum TG,TC,AST,ALT were significantly increased,with significant statistical significance(P<0.01).Liver tissue pathology shows cell steatosis,the volume becomes larger,circular vacuoles of different sizes scattered randomly in the cytoplasm,nucleus is squeezed to one side,and some inflammatory cell infiltration and spot necrosis are obvious performance of steatohepatitis.(2)Compared with the model group,the liver index,serum TG,TC,AST,and ALT of the rats in the high、low-dose,and western medicine groups of Yinqin Qinggan Decoction significantly improved,with statistical significance(P<0.01).The steatosis of hepatic cells was improved to varying degrees,and the improvement of high-dose group was better than that of low-dose group and western medicine group.(3)Compared with the western medicine group,the liver index,serum TG,TC,AST,and ALT in the high-dose Yinqin Qinggan decoction groups were improved,and some of them were statistically significant(P<0.01 or P<0.05).[Research conclusion](1)Compared with normal rats,rats with high-fat diet induced abnormal changes in the morphology of the liver,blood lipids,and liver function,suggesting that a high-fat diet may play a key role in the occurrence of NASH in rats.(2)Yinqin Qinggan decoction has a protective effect on rat liver,which can reduce the content of TG,TC,AST,ALT in the serum of NASH model rats,and improve the pathological state of liver tissue,suggesting that Yinqin Qinggan Decoction may reduce blood lipids and aminotransferases.The activity thus protects the liver from NASH’s chronic liver damage and inflammation.
