| Modified XiaoChaihu Decoction is composed on the basis of the classical prescription"Xiaochaihu Decoction"adding Dried ginger,Coptis chinensis,Fried atractylodes and Poria cocos.It has the function of relieving liver and stomach,regulating the spleen and stomach,acrid opening and bitter descending.Treating spleen and stomach disease.This prescription is used in the treatment of gastric ulcer and chronic gastritis in long-term clinical practice by chief physician Zhou Aijun of Dongguan traditional Chinese medicine hospital.The curative effect is remarkable,which is superior to the original prescription.In the period study,we systematically studied the effective components of the modified Xiao Chaihu Decoction,clarified the effective substances of the prescription and why they’re effective,explained its pharmacodynamic material basis and made it into Modified Xiao Chaihu granules hospital preparation.On the basis of previous studies,the pharmacokinetics and distribution of the main effective components of Modified Xiao Chaihu granules were studied.Objective:In order to provide a scientific basis for the clinical dosage of Modified Xiao Chaihu granules,we studied the pharmacokinetics in rat plasma and the tissue distribution of the main active components of Modified Xiao Chaihu granules.Methods:The mixture of 8 main effective components,such as saikosaponin a,saikosaponin d,glycyrrhizic acid,berberine hydrochloride was used as the test product,and the content of the above effective components were determined by UPLC-MS/MS method.The gastric ulcer model rats and normal rats were given Modified Xiao Chaihu granules by intragastric administration of Modified Xiao Chaihu granules respectively,The plasma pharmacokinetics of the main effective components of the preparation were studied by UPLC-MS/MS method,and the content of the main effective components in the distribution of gastric ulcer model rats and normal rats was compared with the above analysis method.Results:The analysis of the main effective components of Modified Xiao Chaihu granules showed that its linear relationship,precision,accuracy and stability were all conforms to the requirements.The results of plasma pharmacokinetics of the main effective components of the preparation showed that the peak time(Tmax)of these effective components in gastric ulcer model rats were later and the peak concentration(Cmax)were higher than that of the normal rats.The area under the plasma concentration time curve(AUC)and the average retention time(MRT)of the above 8 components were all higher while the clearance rate(CL/F)were less than that of the normal rats.The comparative study on the distribution ofthe effective components in liver,spleen,stomach,kidney and small intestine of the model rats and normal rats showed that all those effective components were higher in model rats compared with normal rats.Among them,berberine hydrochloride,baicalin,liquiritin,glycyrrhizic acid were mainly distributed in the stomach and small intestine,and the distribution is stable.Conclusion:This thesis has achieved the anticipated target,the pharmacokinetic study of the main effective components of Modified Xiao Chaihu granules preparation provided scientific basis for its clinical dosage;The comparative study of tissue distribution of the main effective components in gastric ulcer model rats and normal rats provided scientific experimental evidence for the efficacy of the preparation and laid a foundation for the further study of the mechanism of anti gastric ulcer. |
