Objective:To investigate the dose effect relationship at different moxibustion temperatures and the embodiment of TRPV1 mechanism in the spinal cords,so as to provide a reference for clinical selection of moxibustion temperature.Method:Theoretical part:The dose effect relationship was cleared up and analyzed from the perspective of moxibustion quantity,moxibustion effect and their relationship,based on the ancient and modern literature;the TRPV1 mechanism with moxibustion effect was discussed on the basis of biological characteristics of TRPV1.Experimental part:Totally 40 healthy male SD rats were chosen and stimulated with moxibustion at Zusanli in 41℃,45 ℃ and 49 ℃.Extracellular recording techniques of glass microelectrode was used in the rats’ L4dorsal horn neurons.Capsaicin vehicleand solution was injected into the rats through Zusanlito filter the sensitive neuron to TRPV1.The response of sensitive neurons to TRP V1 in dorsal horn under moxibustion of three different different temperatureswas recorded and compared.Result:Theoretical part:The traditional moxibustion quantity included the "method of making fire,size of moxa cone,cone number and operation method."While,in the modern research,the moxibustion temperature,moxibustion time and the moxibustion temperature-moxibustion time curve were used to summarize the essence of moxibustion quantity.And for the warming property of moxibustion,warming and promoting blood circulation as well as warming and nourishing Qi and blood could be produced.Namely,it had the effect of warming-dredging and warming-reinforcing.And,the production and maximization of moxibustion effect corresponded to a certain amount of moxibustion quantity.Namely,in the moxibustion therapy,there was a dose effect relationship combined with different parameters,among which the moxibustion temperature was the more important factor.Clinically,moxibustion could stimulate the temperature which accorded with the activation characteristics of TRPV1.Therefore,the TRPV1 mechanism,which was in the moxibustion therapy,existed in the skin afferent links,central modulation links,and effect links of tissues and organs as well as other channels and pathways.Among them,the skin afferent link and central modulation link could reflect the moxibustion quantity.Experimental part:The TRPV1 sensitive neurons responded to all the three temperatures.Viewing from the nature of the responding neurons,the neuron in 45℃and 49 ℃ mainly performed in agitation,while the number of agitated and inhibitory neurons appeared same in 41 ℃.Viewing from the character of the responding neurons,different temperatures correspond to differential discharge frequency:41℃-(8.26±2.14)spikes/s,45℃-(4.69±2.61)spikes/s,49℃-(10.66±5.97)spikes/s.Viewing from the change of discharge frequency,the neurons in 41 ℃ was found without obvious change,while the neurons in 45 ℃ and 49 ℃ discharge frequency apparently increased(P<0.01).Conclusion:Theoretical part:Warming is the most important motivating feature of moxibustion.warm-dredging and warm-reinforcing are the most important effect features of moxibustion,and temperature is the key indicators of moxibustion quantity.Moxibustion at a specific temperature can activate TRPV1 and exert the therapeutic effect through various pathways,which is the mechanism of moxibustion dose effect relationship.Experimental part:Sensitive neurons in TRPV1 in spinal cord can response to 41 ℃,45 ℃and 49 ℃,whose character,nature and degree are related to the temperature gradient.
