| AIM:To investigate the effect of Tongbian decoction of AQP3 and AQP4 in slow transit constipation rats via MAPK signal pathway.METHODS:1.Establish a model of slow transit constipation rats.60 SD rats were used in this study,of which 20 were randomly divided into normal roup,and 40 were used to induce STC by feeding with compound diphenoxylate,and the dosage was 8mg/kg/d.After 120 days,10 rats in normal group and 10 rats in model group were sacrificed to detect the indicators to verify that the STC model replicated successfully.2.Experimental research.The remaining rats were divided into normal group,model group,Tongbian decoction group,Tongbian decoction+U0126 group,and given corresponding interventions.After 4 weeks,all rats were sacrificed to detect the indicators.INDICATORS:24-hour number of feces;Fecal water content;the functional transmission function;the numbers of AQP3,AQP4 and MAPKs dectected by Immunohistochemistry,Western Blot and Real-time PCR.RESULTS:1.Compared with normal group,the indexes of rats in the model group were significantly different.The model of STC was established successfully.2.Compared with the model group,the number of feces,the Fecal water content and the functional transmission function were increased,and there was a statistical difference.There was a statistical difference in the number of colon residual feces and in the expression of AQP3 and AQP4.3.The expression of AQP3,AQP4 and MAPK in Tongbian Decoction+U0126 group was lower than that in model group,and was higher than that in normal group.There was a statistical difference.CONCLUSION:1.The slow transit constipation model was successfully replicated,and the compound diphenoxylate was safe and effective,and the model was stable.2.Tongbian decoction can reduce intestinal water absorption and improve the symptoms of STC rats by reducing AQP3 and AQP4.3.Tongbian decoction can treat STC by inhibiting the MAPKs pathway. |
