ObjectiveTo explore the effect of edaravone(ED)on sepsis in ALI and its underlying impact on CC16 protein level in the serum.MethodsAccording to the randomness principle,24 male SD rats were divided into 3groups,the control group(group NS),model group(group LPS)and edaravone treatment group(group ED).Among them,the rats in group LPS and group ED were injected with LPS(10 mg/kg)by tail vein in order to establish ALI models.Correspondingly,the rats in NS group were injected with equal volume of normal saline by tail vein.Then,the rats in ED group were injected edaravone(3mg/kg)immediately after tail vein injection,and normal saline was injected into NS group and LPS group.Eight hours after drug injection,the blood and lung samples were harvested and measured,for example,arterial partial pressure of oxygen in arterial blood gas analysis was measured to calculate of the pulmonary oxygenation index of rats.The contents of malondialdehyde(MDA)and superoxide dismutase(SOD)in lung homogenate were measured by thiobarbituric acid and hydroxylamine method.enzyme-linked immunosorbent assay(ELISA)was applied to detect CC16 protein in the serum.In addition,wet weight and dry weight of lung tissues were measured to calculate the wet and dry weight ratio(W/D),and the lung tissue were made into paraffin sections so that the pathological change of lung tissue would be further observed and evaluated.ResultsIn the LPS group,the content of MDA in the lung tissue,W/D and CC16 protein level in the serum were all significantly higher compared to those in NS group(P<0.01),then SOD content in the lung tissue,arterial oxygen pressure and oxygenation index were lower than that in NS group(P<0.01).In the ED group,the content of MDA in lung tissue,W/D and CC16 protein in serum were all significantly lower than those in the LPS group(P <0.01),then the content of SOD in the lung tissue of ED group,arterial oxygen pressure and oxygenation index were markedly higher than that in the LPS group(P<0.01).Pulmonary histopathological changes: significant changes of lung injury can be seen under the microscope in the LPS group,and the lung injury of ED group is less than the LPS group.ConclusionsEdaravone can weaken acute lung injury in rats induced by LPS :appearance as the changes of arterial oxygen pressure,W/D ratio,MDA and SOD content in lung tissure,lung histopathology.At the same time,it can affect the content of CC16 protein in serum,indicating that CC16 protein may have important significance in the treatment of sepsis lung injury mechanism.
