Background:At present,breast cancer is the most common malignant tumor in female,the incidence is increasingly rising.the incidence accounting for 25% of all malignant tumor,in developed countries about half;The mortality accounting for 15% of cancer deaths,in developed countries about 38%.Scinderin(SCIN)was first founded in chromaffin cells by Bader MF in 1986,the mainly function is meditating polymerization reconciliation,of filamentous actin(F-actin),which affects the cytoskeleton refactoring,resulting in cell migration and movement.In recent years,studies have shown that SCIN involved in the occurrence and development of a wide variety of tumor.In megakaryocytic leukemia,SCIN inhibit proliferation and formation;In gastric cancer,lung cancer,prostate cancer,bladder cancer,SCIN promote the proliferation and metastasis of tumor.But there is rarely reported in breast cancer,this study was to explore the function SCIN in breast cancer.Objective:1.This study was designed to explore the differences expression of SCIN between breast cancer tissue and adjacent to carcinoma.In addition,to discuss the differences expression with different molecular subtypes of breast cancer tissue in the relationship between various clinical factors and breast cancer.2.To construct SCIN-shRNA knockdown lentiviral vector,and transfered tinto MCF-7 cell.Then observe the function of MCF-7 which silencing SCIN.Methods:1.Using immunohistochemical staining method to detect the expression of SCIN in 120 cases of breast carcinoma and 30 adjacent tissue,and comparative analysis SCIN expression in breast cancer tissue and breast cancer molecular subtypes,pathologic stage,age,tumor size,lymph node metastasis in patients and clinicalstage.2.SCIN-shRNA lentiviral vector transfered into MCF-7 cell.3.Western bloting was used to evaluating the efficiency of silencing.4.The wound healing experiment was used to detecting the migration between Lv-shSCIN and Lv-shCON.5.Transwell was used to detecting the invasion between Lv-shSCIN and Lv-shCON.6.CCK8 was used to detecting the proliferation between Lv-shSCIN and Lv-shCON.Results:1.The level of SCIN expression in breast cancer tissue below the tissue adjacent to carcinoma(P < 0.05).SCIN expression is associated with breast cancer molecular subtypes and lymph node metastasis(P < 0.05),irrelevant with age,tumor size,histological grade,clinical stage(P > 0.05).2.The lentiviral vector carry green fluorescence protein gene,under the fluorescent microscope,the successful transfered cells can observed green fluorescence.The Lv-shCON group and Lv-shSCIN group,green fluorescent area more than 90% and 85% respectively,prove the transfection succeed.3.Western bloting experiment result show that campared with Lv-shCON,Lv-shSCIN group the SCIN protein level was lower,the difference was statistical significance(p<0.001).4.The wound healing experiment show that the ablity of wound healing in Lv-shSCIN group was stronger than Lv-shCON,The result indicate that silencing SCIN can promote migration in MCF-7 cell.the difference was statistical significance(p<0.01 and P<0.05).5.Transwell show that in Lv-shSCIN group,the amount of cell pass through the transwell cellar more than Lv-shCON.The result indicate that silencing SCIN can promote the ability of invasion in MCF-7 cell.the difference was statistical significance(p<0.05).6.CCK8 show that silencing SCIN can promote the ability of proliferation inMCF-7 cell,the difference have statistical significance from the third day(P<0.05?P<0.01?P<0.001?P<0.0001).Conclusions1.The expression of SCIN in breast cancer tissues was lower than those of adjacent,associated with breast cancer molecular subtypes and lymph node metastasis.2.This research succeed silencing SICN in MCF-7 cell,the proliferation ?invasion and migration ability are enhanced obviously.
