Objective: To investigate the correlationship among 25-hydroxyvitamin D and oxidative stress and DNA damage in patients with lung squamous carcinoma,and to research serum 25-hydroxyvitamin D whether there is a protective role in patients with lung squamous carcinoma.Methods: Selection in April 2016 to April 2017 at respiratory medicine of southwest medical university first affiliated hospital of 60 patients with lung squamous carcinoma as experimental group,30 normal healthy individuals for the same period served as the normal control group.Enzyme-linked immunosorbent assay(ELISA)to determine the serum 25-hydroxyitamin D level.By xanthine oxidase method and thiobarbituric acid method(TBA)to detect the serum SOD activity and MDA content;Using flow cytometry instrument through dihydro-ethidium(DHE)probe and 2’,7’-dichlorofluorescence(DCFH-DA)probe to detect the peripheral blood mononuclear cells(PBMC)average fluorescence intensity of DHE and DCF.Using flow cytometry to detect γ-H2AX’s average fluorescence intensity and the level of γ-H2 AX in PBMC;And in 25-hydroxyvitamin D and γ-H2 AX level,γ-H2 AX,DHE,DCF average fluorescence intensity,SOD activity and MDA content of correlation analysis.Results: 1.ELISA for determination of serum 25-hydroxyvitamin D levels shows lung squamous carcinoma group’s 25-hydroxyitamin D level(19.42 ± 6.45)ng/ml is lower than normal control group(56.08 ± 15.16)ng/ml(P < 0.05),the difference was statistically significant.2.Xanthine oxidase method to detect the serum SOD activity shows lung squamous carcinoma group’s serum SOD activity(63.41 ± 21.55)U/ml is lower than normal control group(106.09 ± 11.56)U/ml(P < 0.05),the difference was statistically significant.TBA method to detect the serum MDA content shows lung squamous carcinoma group’s serum MDA content(17.94 ± 7.19)nmol/ml is higher than normal control group(5.49 ± 2.41)nmol/ml(P < 0.05),the difference was statistically significant.3.Using flow cytometry instrument to detect the average fluorescence intensity of DHE,DCF shows lung squamous carcinoma group’s DHE average fluorescence intensity(169.52 ± 22.42)is higher than normal control group(96.39 ± 9.51)(P < 0.05),the difference was statistically significant.Lung squamous carcinoma group’s DCF average fluorescence intensity(136.54 ± 16.94)is higher than normal control group(78.04 ± 12.06)(P < 0.05),the difference was statistically significant.4.Using flow cytometry instrument to detect the level of γ-H2 AX and the average fluorescence intensity of γ-H2 AX in PBMC shows lung squamous carcinoma group’s level of γ-H2AX(37.91% ± 16.55%)is higher than normal control group(3.51% ± 1.16%)(P < 0.05),the difference was statistically significant.Lung squamous carcinoma group’s average fluorescence intensity of γ-H2AX(8.74 ± 1.98)is higher than normal control group(3.63 ± 0.60)(P < 0.05),the difference was statistically significant.5.Correlation analysis showed: Lung squamous carcinoma group’s 25-hydroxyvitamin D level and PBMCs’ DHE average fluorescence intensity were negatively correlated(P < 0.05).Lung squamous carcinoma group’s 25-hydroxyvitamin D level and PBMCs’ DCF average fluorescence intensity were negatively correlated(P < 0.05).Lung squamous carcinoma group’s 25-hydroxyvitamin D level and PBMCs’ γ-H2 AX average fluorescence intensity were negatively correlated(P < 0.05).Lung squamous carcinoma group’s 25-hydroxyvitamin D level and PBMCs’ γ-H2 AX levels were negatively correlated(P < 0.05).Lung squamous carcinoma group’s 25-hydroxyvitamin D level and serum MDA content were negatively correlated(P < 0.05).Lung squamous carcinoma group’s 25-hydroxyvitamin D level were positively correlated with serum SOD activity(P < 0.05).Conclusion:1.The lung squamous carcinoma patients with obvious 25-hydroxyvitamin D deficiencies or lack of phenomenon.2.Lung squamous carcinoma patients serum MDA content increased significantly,serum SOD activity decreased obviously,the average fluorescence intensity of DHE and DCF in PBMC increased significantly,lung squamous carcinoma patients’ PBMCs with obvious oxidative stress.3.Lung squamous carcinoma patients PBMCs’ DNA damage rate and γ-H2 AX average fluorescence intensity increased significantly,lung squamous carcinoma patients’ PBMCs with obvious DNA damage in the body.4.Lung squamous carcinoma patients with 25-hydroxyvitamin D levels and negatively correlated with oxidative stress.5.Lung squamous carcinoma patients with 25-hydroxyvitamin D levels and negatively correlated with DNA damage.6.25-hydroxyvitamin D can reduce oxidative stress and DNA damage of lung squamous carcinoma.
