Chapter I Expression of TPX2 in cholangiocarcinoma and itsrelationship with clinicopathological parameters and prognosisObjective : To investigate the expression of targeting protein for xenopus kinesin-like protein 2(TPX2)in cholangiocarcinoma tissue and its clinical significance.Methods : The expression of TPX2 protein was detected by immunoh-istochemistry in 70 cases of cholangiocarcinoma and 4 cases were randomly selected to detect the expression of TPX2 protein by Western blot.The expressions of TPX2 protein and the relationship between the clinicopathological parameters and prognosis of patients.Result:The positive expression rate of TPX2 protein in cholangiocarcinoma tissues(44.3%(31/70)vs.5.7%(3/70),P <0.05)and the expression of West blot were higher than those in paracancerous tissues.The expression of TPX2 protein was correlated with TNM stage,lymph node metastasis and survival(all P <0.05).The postoperative survival rate of TPX2 positive patients was significantly lower than that of negative patients(2-year survival rate 28.1% vs.80.3%;3 Annual survival rate: 14.2% vs.60.7%,all P <0.05).Conclusion:TPX2 protein is highly expressed in cholangiocarcinoma and its expression intensity may be a risk factor for postoperative metastasis and recurrence of cholangiocarcinoma.TPX2 protein may play an important role in the progression from precancerous lesions to cholangiocarcinoma.Intensity may be a potential target for evaluating a potential biological indicator and treatment for early diagnosis and prognosis of cholangiocarcinoma.Chapter II Effect of TPX2 gene on the biological characteristics ofhuman cholangiocarcinoma cell lineObjective:Observe the effect of TPX2-si RNA knockdown on the expression of TPX2 protein in cholangiocarcinoma cells to investigate the correlation between TPX2 gene and cholangiocarcinoma,and provide ideas for exploring TPX2 as a gene therapy target for cholangiocarcinoma.Methods : The effects of TPX2 expression on the proliferation,cell cycle,invasion,migration and apoptosis of cholangiocarcinoma cells were studied by CCK-8,flow cytometry analysis and transwell assay.Cell cycle,apoptosis and EMT-related target protein expression were detected by immunoblotting.Result:Knockdown of TPX2 expression inhibited proliferation,G2-M arrest,apoptosis and inhibited invasion and migration of cholangiocarcinoma cells.Treatment of cholangiocarcinoma cells with TPX2 si RNA resulted in upregulation of cyclin A1,cyclin B1,E-cadherin,p53,and Bax,and downregulation of Ki67,cyclin D1,Bcl-2,N-cadherin,β-cadherin CDK2,MMP-9,and MMP-2.Conclusion : Knockdown of TPX2 expression can induce G2-M arrest of human cholangiocarcinoma cells,promote apoptosis of cholangiocarcinoma cells,inhibit proliferation,invasion,and metastasis of cholangiocarcinoma cells,and may be a potential target for targeted drug therapy of cholangiocarcinoma.
