ObjectiveTo observe the effect of Buyang Huanwu Decoction on the expression of Integrin α5β1 and new vessels in the cortex of cerebral ischemia/reperfusion injury.Methods1.Buyang Huanwu Decoction preparation.2.The middle cerebral artery occlusion reperfusion model in male rats was established by wire suture method.After successful modeling,the rats were randomly divided into sham operation group,model group,Buyang Huanwu Decoction group,Yiqi group and Huoxue group.Then each group was divided into three subgroups on day 4,7 and 14 post-ischemia.The rats were scored according to the modified neurological deficit.Longa five grade scoring method was used to evaluate the extent of neurological impairment in rats.TTC staining was used to determine the changes in infarct size of brain tissue on day 4,7,and 14 after ischemia.Hematoxylin and eosin(HE)staining of brain tissue was observed.The expressions of CD34 and integrin α5β1 in the brain tissue of ischemic rats were detected by immunohistochemistry.The number of positive cells was analyzed,and the microvessel density was compared.Results1.Neurological function score.Compared with the model group,there was no significant difference in the neurological function score among the Buyang Huanwu Decoction group,the Yiqi group and the Huoxue group at day 4,while the difference was significant at day 7 and 14(P<0.05).The Buyang Huanwu Decoction group was significantly lower than the Yiqi group and Huoxue group(P<0.05).2.At 4d,there was no significant difference in infarct volume between the experimental groups(P>0.05).Compared with the model group,the infarct volume decreased significantly(P<0.05)in the Buyang Huanwu Decoction group,the Yiqi group and the Huoxue group at 7d and 14 d.The Buyang Huanwu Decoction group was significantly lower than the Yiqi group and the Huoxue group(P<0.05),and there was no significant difference between the Yiqi group and the Huoxue group(P>0.05).3.Microvessel density at the ischemic side.At 4d,there was no significant difference in microvessel density between the experimental groups.After cerebral ischemia,7d,14 d Yiqi Huoxue group compared with the model group,the cerebral ischemia surrounding tissue neovascularization density increased significantly.Compared with the Yiqi group and the Huoxue group,the Buyang Huanwu Decoction group increased significantly,(P<0.05).4.Expression of integrin α5β1.Integrin expression increased significantly after cerebral ischemia in rats.After cerebral ischemia 4d,the expression of integrin α5β1 in Yiqi Huoxue group was slightly higher than that in model group,but the difference was not statistically significant(P>0.05).Compared with the model group,the expression of integrin α5β1 increased significantly after cerebral ischemia in 7d,14 d and Yiqi Huoxue group(P<0.05).The Buyang Huanwu Decoction group was significantly higher than the Yiqi group and the Huoxue group(P<0.05).ConclusionCerebral ischemia resulted in significant elevation of the density of neovascularization in the ischemic region,associated with significantly increased of Integrin α5β1 expression.Buyang Huanwu Decoction administration significantly improve neurological function and reduce infarct volume,compared with the ischemia group.The expression of Integrin α5β1 was increased and the vascular density was improved,and Buyang Huanwu Decoction administration is an important pathway of angiogenesis.
