| As a kind of nonionic surfactant,nonylphenol is widely used,leading to nonylphenol can be detected in various medias such as groundwater,soil,atmosphere and foods.The nonylphenol can eventually enter human’s body through the food chain,meaning that it is necessary to establish an accurate detection method of nonylphenol.Therefore,in this paper,nonylphenol was purified to candidate reference material by prep-HPLC.The purity of nonylphenol was determined by mass balance method,which provided reference for the detection of nonylphenol samples.Although there are many studies on the reproductive toxicology of nonylphenol,few studies have been carried out by metabonomics.In this paper,the dose-effect relationship of nonylphenol was analyzed by the serum metabolic group,and the possible biomarkers were screened from the metabolites in the serum.The results of the study not only do help to further understand the biochemical effects of nonylphenol,but also provide a reference for the in-depth study of the toxic effect mechanism of nonylphenol.The optimized conditions of prep-HPLC were: concentration of raw material was 30 mg/mL,mobile phase was water(A)and methanol(B)in gradient condition;the wavelength was 278 nm,the flow rate was 12 m L/min,the sampling amount was 2.0 mL.The purity of product was 99.90%,which was improved compared with the raw material purity of 97.97%;the method was stable and good stability for nonylphenol could be kept under 4 degrees in one month.Qualitative analysis was made through GC-MS and IR.Mass balance method was taken to determine the purity was 97.72%,and the overall uncertainty was 2.41%.Secondly,a metabonomics analysis method based on UPLC-Q-TOF/MS was established.Data obtained from metabonomics were analyzed by multivariate statistical analysis and integration analysis.Finally,biomarkers of related endogenous metabolites with toxic effects were found.The metabolic pathways of these 15 biomarkers contained glycerophospholipid metabolism,carbohydrate metabolism,amino acid metabolism,lipid metabolism,which were PC(14:0/18:0),PC(18:0/14:0),PC(16:0/16:0),PC(22:6),PC(18:0/22:6),PC(20:2/20:4),PC(20:3/20:5),PC(22:6/18:2),L-cystathionine,oxoglutaric acid,galactose 1-phosphate,5-aminolevulinic acid,ketoglutaric acid,linoleic acid and taurocholic acid.Through the analysis,the content of PC,L-Cystathionine,Galactose 1-phosphate and 5-Aminolevulinic acid decreased with the increase of exposure time,the content of other substances increased;the biological function of biomarkers indicated that nonylphenol could produce toxic effects on human beings,such as oxidative stress and mitochondrial dysfunction.The experiment was of great significance for toxicity identification and evaluation of nonylphenol. |
