Naringin,a peripheral antitussive,has been found that it has significant antitussive effect and phlegm-eliminating effect without any central inhibition by oral adminitration.Cough belongs to respiratory tract disease of which inhalation drug has significant advantages on therapy because inhalation drug can get into blood directly through pulmonary alveoli.Some of inhalation drug can deposit on respiratory tract where is the target of most of the inhalation drug.At present,there is no antitussive inhalation on market.The aim of this study is to develop the formulation of Naringin.The details were as follows,(1)After screening the solution of Naringin,type and dosage of cosolvent,antioxidant,two kind of solution has been decided to be research for the further study.(2)The Naringin solution for nebulization was prepared and three batches of it were prepared.According to the regulation of ChP(2015),the stress testing were carried out.(3)The particle size distribution measurement of two formulation was carried out by using spraytec and next generation pharmaceutical impactor(NGI).The result of spraytec claim that the percentage of particles which size was between 1~5μ m was 62.9%,and the percentage of particles which size was between 5~20μm was 33.8%.The result of NGI indicate that the FPF%(fine particle fraction)was 64.2±2.4%,and the MMAD(mass median diameter)was 3.908±0.180.The results accord with the regulation of ChP.(4)The cough model of guinea pig was conducted by using citric acid.The effects of low,medium and high dosed of drugs were observed and its result was compared with the effect of levodropropizine.The antitussive effect of low,medium and high dosed of drugs were 21.6±7.5%、33.6±10.4%、50.8±8.6% respectively,and the antitussive effect of levodropropizine was 33.2±7.3%.This result support the importance of the development of Naringin for inhalation.
