In the present study,Yunnan Bream skin is used as experiment materials,and acid-soluble collagen was isolated from bream skin,by low temperature acid method,and the physicochemical properties of collagen in Yunnan Bream skin would be investigated.The starch of pachyrhizus and polyphenols of rambutan that were added to the collagen membrane,the collagen membrane were improved and the technical parameters and theories of the comprehensive utilization of sea Bream and the development of edible film were discussed and studied.Through the analysis of the experimental results,the main conclusions are as follows:1.ASC was demonstrated as type I collagen,and was abundant in Gly,Pro and Ala,whereas low in Tyr,Met,and Cys.The maximum UV absorption peak of ASC was at 230 nm.Fourier transform infrared spectroscopy and X-ray diffraction proved that the ASC molecule was regularly arranged and the helical structure of which was maintained.Scanning electron microscope showed that ASC-B had a three-dimensional micro structure cross linked by collagen fibres and was suitable to be used as the base of biomaterial Peptide analysis showed that the main amino sequence of ASC was Gly-X-Y,which was in agreement with characteristics of primary structure of collagen.When pH<4 and NaCl concentration above 4%,the solubility of ASC was relatively high.2.The collagen film was modified through addition of pachyrhizus starch,and the results showed the water resistance behavior,water vapor permeability,opacity,mechanical strength were significantly improved;Fourier transform infrared spectroscopy showed a strong interaction between collagen molecules and pachyrhizus starch molecules,X-ray diffraction demonstrated that collagen-starch composite film has two-phase region,Scanning electron microscope revealed that the collagen and pachyrhizus starch molecules were compatible and the film matrix was uniformly and regularly distributed;3.Crosslinking modification of blend film was performed through addition of rambutan polyphenols.The results showed that the water resistance behavior,water vapor permeability,opacity,and mechanical strength of the three component film were further improved,whereas the improvement is not obvious;X-ray diffraction and Fourier transform infrared spectroscopy showed show that the ternary composite membrane is more complex in the amorphous region,and there is a strong interaction between rambutan polyphenols,pachyrhizus starch,and collagen.Scanning electron microscope showed that there was good compatibility between rambutan polyphenol,pachyrhizus starch and collagen.The composite film was still smooth and smooth,and the membrane matrix was uni'form and compact.
