Objective:Objective to study the effect of Danshen injection, Shen Fu Injection and Shenmai injection after myocardial infarction heart failure rat N-terminal pro brain natriuretic peptide(NT-proBNP) precursors, left ventricular ejection ejection fraction(LVEF) and myocardial pathological morphology, to measured syndrome preliminary exploration of TCM Syndrome Types in the rat model of congestive heart failure.Methods:50 SD rats were fed for 1 week in the SPF animal laboratory. 1 week later,according to the random number table method randomly selected 6 as the normal control group, 10 as the sham operation group, and the remaining 34 for operation group. After 8 weeks, using ultrasonic echocardiography detected in rat EF, orbital blood, centrifugal blood obtained from the serum by ELISA method for the detection of NT-proBNP values, according to the clinical diagnosis standard, combined with EF,NT-proBNP identified rat models of heart failure. Will become the rat models of heart failure according to the random number table method were randomly divided as Danshen injection group, Shenfu group, Shenmai group, model group, respectively:Danshen injection, Shen Fu injection, Shenmai injection, sterile water for injection,through intraperitoneal administration, the sham operation group were intraperitoneally injection of sterile water for injection. The dosage of 6.0ml/(D / kg),the dosage of Shen Mai injection was the same as that of Shen Mai injection, the dosage of Salvia miltiorrhiza injection was 2.5ml/(D / kg), and 2 weeks of treatment.After the end of the treatment, the EF value of the rats in each group was detected by ultrasound, ELISA method was used to detect the level of NT-proBNP in serum of each group, and the HE staining method was used to detect the pathological changes of myocardium in rats.Results:1. Compared with the normal group, the LVEF value of the model group was lower, the difference was statistically significant(P < 0.01), the serum level of NT-proBNP was significantly increased, and the difference was statistically significant(P < 0.01).2. After administration, Salvia miltiorrhiza group rats were more active, sensitive,fur smooth supple luster, diet, body weight close to the sham operation group.3. After administration, compared with the model group, Danshen injection can improve heart failure rats EF value, the difference has statistical significance(P<0.01);Salvia miltiorrhiza can Shenmai injection can improve heart failure rats FS value, the difference is statistically significant(P<0.01); Salvia miltiorrhiza can reduce the lvids value, the difference was statistically significant. And Shenfu group, Danshen injection can enhance the value of EF in rats with chronic heart failure, the difference has statistical significance(P<0.01); Salvia miltiorrhiza can Shenmai injection can improve heart failure rats FS value, the difference is statistically significant(P<0.01);Salvia miltiorrhiza group can reduce the lvids value, the difference was statistically significant. Compared with Shenmai group, Danshen injection can improve the heart failure rats LVEF value, the difference has statistical significance(P<0.01); Salvia miltiorrhiza can Shenmai injection can improve heart failure rats FS value, the difference is statistically significant(P<0.01); Salvia miltiorrhiza group can reduce the lvids value, the difference was statistically significant.4. After administration, compared with the model group, Danshen injection can reduce the level of NT-proBNP, the difference has statistical significance(P<0.01);and Shenfu group, Danshen injection can reduce the level of NT-ProBNP, the difference has statistical significance(P<0.01); compared with Shenmai group and Dan Shen injection can reduce the level of NT proBNP, the difference has statistical significance(P<0.01).5. Myocardial pathological morphology. Normal group: myocardial structure clear, intercalated disc, stripes and clear, uniform arrangement of muscle fibers, neat,myocardial nuclei were located in the edge, nuclear is normal in size and shape,myocardial fiber no fracture defects and blanks.Model group:large area myocardial infarction. Myocardial cells were intertwined,nonstriated, intercalated disc, muscle fibers arranged disorder, broken and missing,myocardial cell pyknosis, the visible region of the blank, adjacent cells intercalated discs gap widened significantly, most of myocardial necrotic tissue dissolvedabsorption.Salvia miltiorrhiza group: obvious infarction foci. Stripes and intercalated disc is obvious, the muscle fiber arranged mussily, local breakage and loss, myocardial cell pyknosis, showing a small amount of blank area.Shen Fu Group: see obvious infarction foci. Stripes and intercalated disc is not obvious, muscle fibers were arranged in disorder, partial breakage and loss,myocardial cell pyknosis, visible a few gaps in the area, a small amount of cellular edema.Shen Mai group: obvious infarction. Stripes and intercalated disc is not obvious,muscle fibers arranged mussily, local breakage and loss, myocardial cell pyknosis,visible large gaps in the area, a large number of cells edema.Conclusion:Salvia miltiorrhiza injection can increase the value of LVEF and FS in rats with heart failure after myocardial infarction, reduce the value of LVIDS,reduce the level of NT-ProBNP in rats with heart failure after myocardial infarction,improve the degree of myocardial infarction in rats with heart failure after myocardial infarction. According to the theory of "taking square test", the syndrome type of heart failure rat model of myocardial infarction was presumed to form the syndrome of blood stasis syndrome. |
