Objectives:Through feilaokang united isoniazid influenced the lung, spleen tissue of drug-resistant tuberculosis mouse to explore the synergistic effect of feilaokang treatment the drug-resistant TB.Methods:Based on the preliminary experiment research,by tail vein injection isonazid-resistant M.TB suspension to build the isonazid-resistant TB mouse models and injection H37RV standard M.TB suspension to build the standard TB mouse models.There were 120 SPF kunming mouse in the experiment,half of male and female, randomly divided into 6 groups,per group 20. R-INH TB mouse model be divided into four groups, respectively, used 0.9% sodium chloride injection (model group) and 1% isoniazid aqueous solution(isoniazid group),1 g/ml feilaokang water decoction (feilaokang group), feilaokang united isoniazid liquid (feilaokang united isoniazid group) to intervention. Standard TB mouse models (standard strains group) used 0.9% sodium chloride to lavage;Blank group was SPF healthy mouse which did not injection bacteria.Every week weighted once, had continued lavage 56 days,then puted to death.Taked lung,liver and spleen tissue to make pathological examination.Removed part of lung tissue to make bacteria culture.Made bacterium colony count to the M.TB from the culture.Made the M.TB inactivate for separation and purification of DNA.Used the method of whole genome sequencing to make M.TB DNA analysis.In standard strains (H37RV) as the reference sequence, selected associated with ahpC gene mutations to make a contrastively analyzed.Results:①The mouse in feilaokang united isoniazid group,whiches mental state was well, showed response sensitively, action flexible, Coat glossy,and the nutritional status was good, performance for skin elastic and in color of pink, muscle was plentiful and substantial, physical shape was strong.And the mouse’s weight increasing amplitude was significantly greater than feilaokang group, isoniazid group, model group, standard strains group and blank group (P< 0.05).These mouse of feilaokang group were poor mental state,the reaction and action were a bit slow, coat was matt, and the nutrition was poor, performance for skin empurples, less elastic, normally skin temperature, body slants thin, but there was no frame exposed.The mouse gained Weight was significantly greater than the model group,standard strains group and the blank group (P<0.05).These mouse of isoniazid group was poor mental state,the reaction and action were a bit slow, coat was matt, and the nutrition was not very well, showed for skin empurples, less elastic, normally skin temperature, body slants thin, but there was no frame exposed.Isoniazid group compared with feilaoakng group that The mouse gained weight had no significant difference (P>0.05).These mouse of blank group was good mental state,showed the reaction was responsive, flexible motion, coat glossy.the nutrition was medium,performance for well-built, skin elastic,muscle fruitful.The mouse gained weight of blank group was obviously greater than the model group and standard strain group (P< 0.05).②Feilaokang group compared with model group, the injury degree of lung tissue and spleen tissue was significantly lower (P<0.05).The damage of liver tissue is mitigated, but the difference was not obviously, there was no statistically significant(P>0.05).Isoniazid group compared with model group, the injury degree of lung tissue and spleen tissue was significantly lower (P<0.05).The damage of liver tissue is mitigated, but the difference was not obviously,there was no statistically significant (P>0.05).Feilaokang united isoniazid group compared with model group, the injury degree of lung tissue and liver tissue was significantly lower (P<0.05). The damage degree of spleen tissue is lighter than model group, but there was no statistically significant difference (P>0.05).Feilaokang group compared with isoniazid group, injury degree of lung tissue and liver tissue was significantly lower (P<0.05).The damage degree of the spleen tissue has no obviously difference (P>0.05).Feilaokang group compared with feilaokang united isoniazid group, that the damage degree of the lung tissue, liver tissue and spleen tissue were no obviously difference (P>0.05).Isoniazid proup compared with feilaokang united isoniazid group, that the damage degree of the lung tissue was heavier (P<0.05).The damage degree of the liver tissue has no obviously difference (P>0.05). The damage degree of spleen tissue is lower (P<0.05).③The grinding liquid of the mouse’s lung tissue of feilaokang group which was diluted 10 times and 100 times,cultivate the number of mycobacterium tuberculosis were significantly fewer than model group (P<0.05).The grinding liquid of the mouse’s lung tissue of isoniazid group which was diluted 10 times and 100 times,cultivate the number of mycobacterium tuberculosis were significantly fewer than model group (P<0.05).The grinding liquid of the mouse’s lung tissue of feilaokang united isoniazid group which was diluted 10 times and 100 times,cultivate the number of mycobacterium tuberculosis were significantly fewer than model group (P<0.05).The grinding liquid of the mouse’s lung tissue of feilaokang group which was diluted 10 times and 100 times,cultivate the number of mycobacterium tuberculosis were no statistical differences compared with model group (P<0.05).The grinding liquid of the mouse’s lung tissue of feilaokang group which was diluted 10 times and 100 times,cultivate the number of mycobacterium tuberculosis were significantly more than feilaokang unnited isoniazid group (P<0.05).Isoniazid group compared with feilaokang united isoniazid group,the grinding liquid of the mouse’s lung tissue was diluted 10 times which cultivated the number of mycobacterium tuberculosis was no statistical differences(P<0.05).the grinding liquid of the mouse’s lung tissue was diluted 100 times which cultivated the number of mycobacterium tuberculosis was significantly more(P<0.05).④Seven bases of ahpC gene corresponding codon and amino acid had not been changed. The feilaokang group and feilaokang combine isoniazid group 2729621 of gene mutation locus, the corresponding codon Gcc changed to Tcc, by alanine to serine.Conclusion: ① used the method of mouse tail vein injection to establish the resistant-isoniazid TB mouse models and standard TB mouse models were succeed.② Feilaokang could significantly increase the growth of weight (P< 0.05), and improved the mental state and nutritional status in mouse compared with model group.③ Feilaokang could improve isoniazid ease the pathological damage degree of the lung tissue, liver tissue and spleen tissue,in which relieved the pathological damage of lung tissue and spleen tissue was obviously (P< 0.05)④Feilaokang water decoction, isoniazid aqueous solution and feilaokang united isoniazid liquid has the function of inhibiting the growth of mycobacterium tuberculosis in the lung tissue of mouse, which inhibited the lung tissue grinding liquid that diluted 10 times cultivated MTB was significant(P< 0.05).
