| Objective : To investigate the effect of electroacupuncture(EA)at “Zusanli(ST36)” etc.on the general symptoms,sugar level,gastric emptying,electrogastrogram(EGG)and expression of Ghrelin m RNA and SCF/c-kit in the gastric antrum in diabetic gastroparesis(DGP)rats.So as to explore its mechanism underlying improvement of diabetic gastroparesis,and comparing the different between the effect of EA acupoint and medication.Methods: The DGP model was established by a single-dose intraperitoneal injected of streptozotocin(2%,55mg/kg)and raised by high-sugar and high-fat fodder 8 week.Rats were randomized into the normal control,DGP model,EA acupoint,EA non-acupoint and medication(metoclopramide)groups(n=10 rat/group).The blood sugar level was measured by glucometer,the effects on gastric emptying were measured after intragastric application of phenol red.The EGG was measured by BL-420 F.The expression of Ghrelin m RNA and SCF m RNA of gsatric antrum were examined by quantitative real-time PCR.The expression of c-kit of gsatric antrum were examined by En vis ion imm uno his tochem is try.Results : 1)Compared with the normal control group,symptoms integral and blood sugar in rats in the model group,EA acupoint group and metoclopramide group were significantly higher than those in normal control group(P < 0.01).The gastric emptying and small intestinal propulsion rates in the model group were significantly decreased(P < 0.01).Compared with the model group,The blood sugar in the EA acupoint group were significantly decreased(P < 0.05).The gastric emptying and small intestinal propulsion rates in the EA acupoint group and metoclopramide group were significantly higher than those in model group(P <0.05 or P<0.01).2)The each group dominant frequency(DF)had no significant difference(P>0.05).Compared with the normal control group,The normal gastric slow wave percentage and dominant power(DP)in the model group were significantly decreased(P < 0.05).Compared with the model group,the normal gastric slow wave percentage and DP in the EA acupiont group and metoclopramide group were significantly increased(P<0.05).3)Compared with the normal control group,the expression of antrum Ghrelin m RNA increase and SCF m RNA decrease in the model group,EA acupoint group and metoclopramide group were significantly variation than those in normal control group(P<0.01).Compared with the model group,EA acupoint group and metoclopramide group could down regulate the expression of Ghrelin m RNA(P<0.01),up-regulate the expression of SCF m RNA(P<0.01,P<0.05).4)Through the light microscope,we can find the expression of c-kit lie between shades of blue in the gastric antrum,they were fusiform shape or irregular edges.The c-kit positive cells of normal were larger quantities than that of the model group,the latter ’s significantly reduced and scattered.Compared with model group,the c-kit positive cells number of EA group and metoclopramide group were much more than that of the model group(P<0.05).Conclusion: 1)Combination of streptozotocin and high-sugar and high-fat irregular diets could induce DGP rat model successfully.2)EA at ST36,ST21 and SP6 can improve the general symptoms,blood sugar,gastric emptying and gastric electricity,promoted the gastrointestinal motility of DGP rats,which is closely associated with its effects in regulating the expression of ghrelin m RNA and SCF/c-kit in gastric antrum.3)The effect of improving gastric emptying is comparable to metoclopramide. |
