Objective Study on the three parameter Bupleurum capsule serum intervention of bone marrow mesenchymal stem cells in the intervention effect of P38 MAPK pathway differentiation into cardiomyocytes,observe stem cell proliferation and differentiation,the expression of P38 MAPK protein,cardiac transcription factor MEF2 C,characteristics of myocardial markers CTn T in the differentiation process of evidence,bupleurum three ginseng capsule containing serum on stem cells the intervention effect of myocardial cell differentiation,clinical medicine Chai Husan ginseng capsule is widely used to lay the theoretical foundation and provide more experimental evidence.Methods The first part: the SPF level of 20 healthy rats,male and female were randomly divided into 2 groups,10 rats in each group,the 1 group was the blank plasma group,1 groups of plasma containing groups to basal diet 20 d,21d gavage,medicated plasma group were given Chaihu three ginseng capsule orally,saline blank plasma group treated with intragastric administration of 28 d,the drug at the end of an hour after the abdominal aortic blood centrifugal serum,serum and blank serum produced.The P1 generation of bone marrow mesenchymal stem cells were cultured and passaged to P3 generation,Chai Husan joined the ginseng capsule containing serum in different concentration ratio,compared with the control group,MTT method of Bupleurum three ginseng capsule optimal dosing concentration and selection and dosing time,measured stem cell proliferation.The second part: according to the different treatment factors of bone marrow mesenchymal stem cells were divided into 4 groups: control group,BMP-2 group,bupleurum three inducer ginseng capsule medicated serum +BMP-2 inducer group,bupleurum three ginseng capsule containing serum group,blank serum,were added to BMP-2,bupleurum three ginseng capsule containing serum +BMP-2 and Bupleurum three ginseng capsule containing serum,detected by Westernblotting method.The expression of P38 MAPK protein expression level detection PT-PCR method after MEF2 C intervention,CTn T.Results The results of MTT showed that the concentration of 24 h intervention Bupleurum three ginseng capsule containing serum 20%,showed a large amount of dense BMSCs suspended or deposited on the bottom of Petri dish wall,most in the proliferation of serum containing 20% concentration,to induce 24 h after reaching the maximum density.The results of Westernblotting showed that compared with the other groups,bupleurum three ginseng capsule group +BMP,P38 MAPKm RNA and protein expression was significantly up-regulated(P<0.01);with the best concentration is 20% to three plasma containing radix ginseng capsule +BMP group,bupleurum three ginseng capsule group,control group,BMP group,P38 group of three Radix ginseng capsule MAPKm RNA and protein expression in +BMP group were lower three chai(P<0.05 or P<0.01),and blank group protein expression compared with the other groups were not statistically significant.The results showed that: BMP-2 group,PCR group,BMP-2+ group of serum containing drug serum MEF2 C and the expression level of CTn T was higher than the blank serum group after induction,the difference was statistically significant(P < 0.01);the expression level of BMP-2+ in serum was higher than that of BMP-2 group and medicated serum group(P < 0.05);BMP-2 group and drug serum group the difference was not statistically significant.(P > 0.05)Conclution The medicated serum of Radix Bupleuri three capsule can promote the proliferation of bone marrow mesenchymal stem cells(MSCs)in certain conditions,and promote the differentiation of bone marrow mesenchymal stem cells(BMSCs)into cardiomyocytes by P38 MAPK pathway.
