| Objective:On the treatment of pulmonary fibrosis from attending in TCM clinical experts through summarizing ancient books and clinical practice of the consensus.While the experimental study of maimengdong and gancaoganjiang decoction about lung function has not been reported.The intervention effect of two decoctions on pulmonary fibrosis in rats was systematically studied,we want to compare two prescriptions on pulmonary function in rats with experimental pulmonary fibrosis and related molecular mechanism.To explore the effect on the treatment of experimental atrophic lung disease and the possible mechanism,provide the pharmacological basis and new ideas for clinical medication.Method:In this experiment,80 SD rats were treated with intratracheal injection of bleomycin 5mg/kg.Eighty SD rats were randomly assigned:the normal control group,pulmonary fibrosis model group,the Maimendong of high dose(30g/kg),middle dose(15g/kg),low dose(7.5g/kg),and the gancaoganjiang soup of high dose(15g/kg),middle dose(7.5g/kg),low does(3.75g/kg).After modeling,weigh them for a week in timing.The rats in the model group were treated with normal saline,and the medication group was given the corresponding medicine,once a day,once 1ml/100g after 14 days.(1)The lung function indexes of rats were measured:forced vital capacity(FVC),pulmonary dynamic compliance(Cdyn),0.4 second rate(FEV0.4/FVC%),peak expiratory flow(PEF),maximum mid expiratory flow(MMF)and so on(2)The arterial partial pressure of oxygen,carbon dioxide partial pressure,blood oxygen saturation and so on were measured.(3)Pathological changes of lung tissue were observed by HE,Masson and Sirius red staining in each group.(4)Immunohistochemical method was used to detect the expression of angiogenesis related factors VEGF,CD31 and factor VIII in lung tissues.Results:(1)The analysis of viscera quality and organ coefficient:compared with the normal group,the weight of rats in model group decreased significantly(P<0.05).In the model group,the lung mass and lung coefficient increased significantly due to inflammation and extracellular matrix deposition,Maimendong decoction can reduce lung lesions and lung coefficient of quality(P<0.05).(2)The analysis of pulmonary function:Compared with the normal group,the PEF and MMF were both decreased in the model group(P<0.05),and indicate the airway obstruction.FVC was significantly lower than the normal group,with a decline more than 80%(clinical reference),while the 0.4 second rate(FEV0.4/FVC%)increased(P<0.05),what suggested that bleomycin induced pulmonary fibrosis is a restrictive obstructive pulmonary dysfunction.Maimendong decoction and Gancaoganjiang decoction can improve the animal model of FVC(P<0.05).The group decreased the 0.4 second rate of model animals(P<0.05),and improved the airway resistance(P<0.05).(3)The analysis of blood gas:compared with the normal group,the abdominal aortic partial pressure of oxygen(PO2)and oxygen saturation(SO2)decreased in the model group(P<0.05).There was no difference between the two groups in the blood gas results,so the results of each group were compared:gancaoganjiang decoction can improve PO2 and S02(P<0.05).It is only Maimendongtang decoction overall increase of PO2,there was no significant difference between the model group and this group in SO2.(4)Pathological findings:The lung tissue of fibrotic rats can be divided into fibrotic and relative non fibrotic areas.In the normal group,the alveolar structure was complete and orderly.In the model group,the proliferation of interstitial fibroblasts and the enlargement of alveolar space were observed.Dose group of pulmonary interstitial fibroblasts were filled and red cell distribution in reducing Maimendong decoction.The low dose group of gancaoganjiang decoction decreased in pulmonary interstitial fibroblasts.Sirius red staining result:there was a small amount of collagen in normal group,the distribution of collagen were distributed in the model group.There was statistical difference between the two groups(P<0.05).The high Maimendong Decoction.the middle Maimendong Decoction.the high gancaoganjiang Decoction、the middle gancaoganjiang Decoction can reduce the distribution of collagen and the content of collagen.(5)Immunohistochemical results:The expression of VEGF in normal group was less than that in model group,and there were a large number of VEGF expressions in the model group.There was significant difference between the two groups(P<0.05).Expression and new Maimendong soup and liquorice ginger decoction high dose group could reduce the area of consolidation of the animal model of VEGF microvessel density(P<0.05).The high dose group of Glycyrrhiza uralensis could further reduce the expression of VEGF and new microvessels in the non solid areas of lung tissue of model rats,and increase the number of functional vessels.Conclusion:Maimendong and Gancaoganjiang Decoction can improve the pulmonary fibrosis of rat induced by bleomycin.The former mainly through the improvement of lung function to reduce pulmonary fibrosis,The latter may improve pulmonary fibrosis by inhibiting abnormal pulmonary Blood gas. |
