Objective:This paper studied a new synthesized compound with anti-inflammatory activity based on the combination principles----Blf-ADT.Preliminary studies showed that it has anti-cancer activity.To further study the anti-tumor effect of Blf-ADT,we investigated the mechanism of Blf-ADT in inducing apoptosis of Hela cells.Methods:The effect of Blf-ADT on Hela cells was investigated by Morphological observation,MTT assay and Colony formation assay,respectively.We used Scratch experiments of cells to detect whether Blf-ADT could effect the migration of Hela cells.In order to further study the effect of Blf-ADT on Hela cells apoptosis,Flow cytometry analysis was used to quantify apoptotic cells after treatment with different concentrations of Blf-ADT.At the same time,Western blot analysis was performed to investigate the expression of cell apoptosis related proteins,MAPKs signaling pathway proteins and COX-2.Result:The morphology of treated cells became rounded,detached from the culture flask and suspended in the liquid culture under the inverted light microscope.We found that the inhibition of Blf-ADT was dose-dependent with drug concentration,and time dependent with incubation time.Likewise,the cell migration was reduced when concentration of Blf-ADT increased.The percentage of apoptotic cells was increased from 4.27%to 64.22%.A trend of the gradual rising on apoptosis ratio with drug concentration was observed.The result of Western blot showed that with the increase of treated Blf-ADT concentrations,the cell apoptosis related proteins procaspase-3 and Bcl-2 were decreased significantly,while Bax had no change.Furthermore,the expressions of MAPKs protein p-p38 and p-Erkl/2 were decreased,while p38 and Erkl/2 had no change.On the other hand,we have found that Blf-ADT could decrease the expression of COX-2.Conclusion:Our findings strongly indicate the inhibitory effect of Blf-ADT on the proliferation of the human cervical carcinoma(Hela)cells.The mechanisms may include:induction of apoptosis,inhibition of proliferation and the expression of COX-2,activation of cell apoptosis related proteins.Taken together our data indicate that Blf-ADT is a potential compound which may fight against the human cervical carcinoma.
