| Purpose:Through the observation of Qingzhi Tongmai Granule on rabbit atherosclerosis(AS)metabolism and cholesterol reverse transport receptor SR-BI mRNA and protein expression levels,and to explore the Qingzhi Tongmai Granule mechanism of prevention and treatment of atherosclerosis.Material and method: 1.This experiment selects the experimental animal for healthy male rabbits in the experiment group were taken to the animal method of random number table,we will be 22 rabbits,6 rabbits were randomly selected as control group,the remaining 16 rats were model group.The rabbits of each group every day free drinking water,room temperature maintained at 20 ℃ or25 ℃,after adaptive breeding for one week,the model group model,the modeling method of atherosclerosis model using the traditional high fat diet with vitamin D3 injection replication.The model group injected vitamin D3 into the abdominal cavity once,and the dosage was(600 thousand,IU/kg).The blank group was given normal feeding pellets,and the same amount of normal saline was injected on the first day after the experiment.High fat diet for 4 weeks,from the blank group and the model group were randomly selected each rabbit carotid artery pathological section,using HE to observe the pathological changes of carotid plaque staining,the evaluation model is successful or not.After the success of the model,according to the random number table method model of 15 rabbits,with 5 rats in each group were randomly divided into model group,simvastatin group(Western medicine group),Qingzhi Tongmai granule group(Chinese medicine group)and then began to simvastatin group(Western medicine group),Qingzhi Tongmai granule group(Chinese medicine group)drug treatment,two groups were once daily oral intragastric administration of simvastatin,Qingzhi Tongmai Granules,the same continuous intragastric administration for 4 weeks.2.To the eighth weekend of fasting,all rabbits were treated with ear vein blood samples,separation of serum total cholesterol,using automatic biochemical analyzer detected in rabbit serum(TC),triglyceride(TG),low density lipoprotein(LDL-C)and high density lipoprotein (HDL-C)content were detected in rabbits oxidized low density lipoprotein(OX-LDL)levels by enzyme-linked immunosorbent assay(ELISA).To the 9 weekend of fasting by ear vein anesthesia after the death of animal,open abdomen,liver caudate lobe,the expression level by RT-PCR method to detect the rabbit liver tissue SR-BI mRNA;SR-BI protein was detected by Western-blot in rabbit liver tissues expression.3.Using SPSS17.0 software for statistical analysis of 3 groups of experimental data,the measurement data to mean + standard deviation((?) ±S)said that the number of groups were compared with single factor analysis of variance,P<0.01,with statistical significance,P>0.05 was not statistically significant.Results: 1.Four blood test results show that: Compared with the blank group,the content of the model group,the western medicine group,the Chinese medicine group TG,TC,and the LDL-C were all raised(P < 0.01).Compared with the model group,the Chinese medicine group,TG,TC,and LDL-C(P < 0.01)were lower.Compared with the western medicine group,the levels of TG,TC,and LDL-C were all reduced(P > 0.05).Compared with the blank group,the model group,the western medicine group and the Chinese traditional Chinese medicine group had lower HDL-C(P < 0.01).Compared with the model group,the level of HDL-C in western medicine group and Chinese medicine group increased(P < 0.01).Compared with the western medicine group,the Chinese traditional Chinese medicine group had lower HDL-C(P > 0.05).2.Results of oxidized low density lipoprotein(Ox-LDL): Compared with the blank group,the content of the model group,the western medicine group,and the Chinese traditional Chinese medicine group(P < 0.01).Compared with the model group,the content of the western medicine group and the Chinese traditional Chinese medicine group(P < 0.01)were lower.Compared with the western medicine group,the amount of Ox-LDL(P >0.05)was higher in the Chinese medicine group.3.The results of Western-blot showed that the target band was located at about 36 k Da,and the results showed that the expression of SR-BI decreased in the model group,western medicine group and Chinese medicine group(P <0.01).Compared with the model group,the expression of SR-BI in the western medicine group and the traditional Chinese medicine group(P< 0.01). Compared with western medicine group,there was no significant difference(P>0.05).5.The results of RT-PCR showed that the target band was located at about 457 bp,and the results showed that the expression of SR-BImRNA decreased in the model group,western medicine group and Chinese medicine group(P <0.01).Compared with the model group,the expression of SR-BImRNA in the western medicine group and the traditional Chinese medicine group(P< 0.01).Compared with western medicine group,there was no significant difference(P>0.05).Conclusion: 1.The rabbits were fed with high fat diet.It was found that the contents of TC,TG,LDL-C and OX-LDL were significantly increased in the model group,the content of which was significantly lower than that of HDL-C.2.The expression of SR-BI mRNA in liver tissue of rabbits with atherosclerosis model was increased by Qing Zhi Tong Mai granule.3.The expression of SR-BI protein in liver tissue of rabbits with atherosclerosis model was increased by Qing Zhi Tong Mai granule.4.Through the up regulation of the expression of SR-BI mRNA and protein in the liver cells,it can reverse the cholesterol.5.The increased expression of SR-BI mRNA and protein in the liver cells,and then promote the reversal of cholesterol may be another mechanism for the prevention and treatment of atherosclerosis. |
