A Comparative Study On The Expression Of Fas, FasL And Caspase-8 In The Spinal Cord Of Rats With Sciatic Nerve Injury By Acupuncture And Cerclage

Purpose:This experiment aims at the study of acupuncture at Huantiao point and Jiaji point of sciatic nerve injury damage model of resistance to apoptosis in rats.With immunohistochemistry and Western blot detection method were observed in rat ganglion of Fas and Fas L and Caspase-8protein expression,and then discusses acupuncture treatment resistance to apoptosis in rat sciatic nerve injury.Compare acupuncture Huantiao point and Jiaji point abtiapoptptic effect to the celld of sciatic nerve injury of differences.Material and method:48 SD healthy rats,male and female each are,are divided into four groups according to the random allocation methods:blank group,model group,Huantiao group,Jiaji group,each group of 12.Choose a modified clamps method as the experiment building,made of acute sciatic nerve injury in rats.After the building of the 8th day,the rat of blank group and model group are fixed on the rat holder,15 min each time,1 times a day for 14 days.Huantiao group and Jiaji group acupuncture Huantiao point and Jiaji point respectively,the treatment of blank group and model group is the same to blank group and model group.After treatments for 14 days,take the sciatic nerve clamps around below 2 mm sciatic ganglion ipsilateral L4-L5 segmental spinal cord,Observe HE staining of the rat sciatic nerve,using Western Blot detection method to observe the apoptosis of rat factors of Fas and Fas L and Caspase-8,according to the results of statistical analysis.Results:1.SFI Building after 8 day,compared with the blank group,sciatic nerve injury the SFI of each group is markedly lower than the blank group,with statistical significance(p<0.05),description of motor function decline in rats and the building success.Building after 22 day,compared with model group,Huantiao group and Jiaji group of motor function rise in fats,with statistical significance(p<0.05),and Huantiao group rats sciatic nerve recovery is better than the Jiaji group,with statistical significance(p<0.05).Illustrate that acupuncture can promote neural functional recovery.2.HE The change of rat sciatic nerve damaged HE staining.Sciatic nerve fiber distribution in blank group,myelin uniform distribution,the axon good continuity.Model group of nerve fiber distribution is irregular,myelin sheath and axon degeneration.Compared with model group,Huantiao group and Jiaji group nerve fibers change relatively light,and Huantiao group is better than Jiaji group.3.The Western Blot detection3.1Western blot Fas test results Four groups of experiments the Fas protein expression are,(1)compared with the blank group,model group of Fas protein expression levels increase(p < 0.01),Huantiao group and Jiaji group of Fas protein expression significantly raised(p < 0.01);2)compared with model group,Huantiao group and Jiaji group lowered Fas protein expression(p < 0.01).(3)compared with Jiaji group,Huantiao group of Fas protein expression lowered obviously(p < 0.01),Huantiao group and Jiaji group of Fas L protein expression significantly raised(p < 0.01);2)compared with model group,Huantiao group and Jiaji group lowered Fas L protein expression(p < 0.01).(3)compared with Jiaji group,Huantiao group of Fas L protein expression lowered obviously(p <0.01).3.3Western blot Caspase-8 test results Four groups of experiments the Caspase-8 protein expression are,(1)compared with the blank group,model group of Caspase-8 protein expression levels increase(p < 0.01),Huantiao group and Jiaji group of Caspase-8 protein expression significantly raised(p < 0.01);2)compared with model group,Huantiao group and Jiaji group lowered Caspase-8 protein expression(p < 0.01).(3)compared with Jiaji group,Huantiao group of Caspase-8 protein expression lowered Fas protein expression(p < 0.01).(3)compared with Jiaji group,Huantiao group of Fas protein expression lowered obviously(p < 0.01).4.2Fas L immune organized test results Four groups of experiments the Fas L protein expression are,(1)compared with the blank group,model group of Fas L protein expression levels increase(p < 0.01),Huantiao group and Jiaji group of Fas L protein expression significantly raised(p < 0.01);2)compared with model group,Huantiao group and Jiaji group lowered Fas L protein expression(p < 0.01).(3)compared with Jiaji group,Huantiao group of Fas L protein expression lowered obviously(p <0.01).4.3Caspase-8 immune organized test results Four groups of experiments the Caspase-8 protein expression are,(1)compared with the blank group,model group of Caspase-8 protein expression levels increase(p < 0.01),Huantiao group and Jiaji group of Caspase-8 protein expression significantly raised(p < 0.01);2)compared with model group,Huantiao group and Jiaji group lowered Caspase-8 protein expression(p < 0.01).(3)compared with Jiaji group,Huantiao group of Caspase-8 protein expression lowered obviously(p < 0.05).Conclusion:1.Acupuncture at different points,can improve the recovery of sciatic nerve function and Huantiao group is better than the Jijia group.2.The expression of Fas,Fas L and Caspase-8 in spinal dorsal root ganglion neurons may be related to the decrease of Fas,Fas L and Caspase-8 expression after sciatic nerve injury.3.The expression of Fas,Fas L and Caspase-8 in spinal dorsal root ganglion neurons after sciatic nerve crush injury,and Huantiao group is better than the Jijia group.
