ObjectiveThe literature studies have shown that correlation between pulmonary fibrosis and atrophic lung disease,and the therapeutic effect of Astragalus Angelica sinensis on pulmonary fibrosis.The effect of Astragalus angelica extract on the pulmonary fibrosis was studied by experimental study to investigate whether macrophage polarization is related to the therapeutic mechanism of pulmonary fibrosis,and whether it is related to macrophage phenotype,so as to provide a new target for the treatment of pulmonary fibrosis.MethodLiterature research:The relationship between pulmonary fibrosis and the atrophic lung disease was summed up through the related literatures in recent years from the CNKI.In conjunction with the relevant texts of the Inner Canon of Huangdi and the viewpoints of later physicians of the world,to discuss the relationship between atrophic lung disease and Du Pulse,Dai Pulse,Chong Pulse,to provide a new direction for the treatment of atrophic lung disease.Experiment one:Selecting of 105 ICR male mice with SPF grade,they were stochasticly divided into 7 groups,15 in each group,including the normal group,the model group,and five Chinese medicine treatment groups,five groups of optimizing formula a,b,c,d,e.Bleomycin was injected into,the trachea entad between anonymous cartilage to replicate the pulmonary fibrosis model in mice..The lung was taken out on the twenty-first day for HE staining and aniline blue staining,used to detect the content of hydroxyproline,to verify the optimal formula.Experiment two:60 SPF ICR male mice were randomly divided into 4 groups.The same method was used for modeling,and the number of macrophages in alveolar lavage fluid was detected by flow cytometry on the twenty-first day.Experiment three:Preparing the serum containing optimal prescription to act on the RAW264.7 cells,setting up a control group,and adding LPS for the stimulation,the number of different macrophages was detected by flow cytometry.Experiment four:Culturing the cells in the same way as experiment three and RT-PCR was used to detect the expression of phenotypic related genes in macrophages.ResultLiterature research:Pulmonary fibrosis can be attributed to traditional Chinese medicine Feiwei category.Atrophic lung disease is closely linked with Du Pulse,Dai Pulse,Chong Pulse.The occurrence and development of atrophic lung disease would be influenced by the three。Experiment one:The results of slice staining showed that the pulmonary fibrosis in the model group was the most serious and the five Chinese medicine treatment groups had a certain degree of reduction and the effect of formula A is the best.Compared with the model group,the content of hydroxyproline in the five groups decreased,and formula A was the most significant,with statistical significance.Experiment two:Compared with the normal group,the number of M1 in the model group decreased and the number of M2 increased.Compared with the model group,the number of M1 in the treatment group increased and the number of M2 decreased,and the difference of M1 was statistically significant.Experiment three:Compared with the control group,the M1 type macrophage increased and the M2 type macrophage decreased in the drug group.The difference was statistically significant.Compared with the control +LPS group,the M1 type macrophage increased and the M2 type macrophage decreased in the drug +LPS group.The difference was statistically significant.Experiment four:Compared with the control group,the expression levels of IRF5,IL-12 and NF-kappa B p65 in the drug group were all increased,IRF5 and IL-12 were significantly different,all of them were statistically significant.The gene expression of IRF4,IL-10 and NF-kappa B P50 were all decreased,and the difference of IRF4 and IL-10 was significant,and all of them were statistically significant.ConclusionThe extract of Astragalus Radix Angelicae can reduce pulmonary fibrosis,and its mechanism may be related to promoting the polarization of macrophages to M1 and inhibiting the polarization of M2.
