| Objective:This study first explores the effect of Intermediate Frequency Micro Current on glioma U251 cells,screening appropriate current related parameters,and proving inhibition of the proliferation of glioma cells by noninvasive therapy with Intermediate Frequency Micro Current.Intermediate Frequency Micro Current combined with temozolomide and elemene to determine the effect of Intermediate Frequency Micro Current on cell cycle and apoptosis.The aim is to develop a new noninvasive physical method for tumor therapy——Intermediate Frequency Micro Current.Methods:1.Effect of Intermediate Frequency Micro Current on U251 cellsUsing U251 cells as the experimental object,according to the single variable method,the frequency of 50kHz,respectively,exerting electric current of 50 mA,70 mA,90 mA,110 mA,130 mA,150 mA,each stimulus 30min,for 3 consecutive days,After the experiment,the survival rate of U251 cells is detected by CCK-8,the effect of Intermediate Frequency Micro Current intervention on glioma cells is evaluated,and the experimental parameters are determined.2.Intermediate Frequency Micro Current combined with elemene on U251 cellsAccording to the.above screening tests,the’ electrical stimulation.parameters were determined as 50 kHz,150 mA and 30 min,and the synergistic effect of intermediate frequency current combined with elemene intervention on U251 cells is discussed.The logarithmic growth phase of human glioma U251 cells are exposed to electrical stimulation,elemene group,elemene combined with electrical stimulation.The cell morphology of U251 is observed after electrical stimulation.Cell viability is detected by CCK-8 assay,and apoptosis/death and cell cycle status of each group are analyzed by flow cytometry.3.Intermediate Frequency Micro Current combined with temozolomide on U251 cellsAccording to screening tests,the electrical stimulation parameters were determined as 50 kHz,150 mA and 30 min,and the synergistic effect of Intermediate Frequency Micro Current combined with temozolomide Injection intervention on U251 cells is discussed.The logarithmic growth phase of human glioma U251 cells are exposed to electrical stimulation,temozolomide group,temozolomide combined with electrical stimulation.The cell morphology of U251 is observed after electrical stimulation.Cell viability is detected by CCK-8,and apoptosis/death and cell cycle status of each group are analyzed by flow cytometry.Results:1.With the increase of electrical stimulation frequency(50 mA-150 mA),the relative proliferation rate of U251 cells show a decreasing trend.The survival rate of each experimental group is statistically different from that of the control group(P<0.05).2.The cell survival rate is(86.49±0.23)%(P<0.01)after the effect of Intermediate Frequency Micro Current(50kHz,150mA,30min)on U251 cells.Intermediate Frequency Micro Current combined with elemene(IC50)interfere with U251 cells,observed the number of cells decreased obviously,the intercellular connection decreased,the floating cell fragments in the culture medium,and the cell morphology is shrinking and smaller.cell survival rate is(36.20± 1.04)%(P<0.01).It can increase the rate of cell apoptosis,including early apoptotic cells and late apoptosis cell number in all cells of the ratio of(68.5 ± 1.65)%(P<0.01);in the cell cycle,the main block of cells in G2/M phase,accounting for(57.09± 1.52)%(P<0.01),but the S phase of cell function is not obvious.3.The survival rate of Intermediate Frequency Micro Current combined with temozolomide(IC50)is(33.28 ±1.67)%(P<0.01).The Intermediate Frequency Micro Current combined with temozolomide increased the apoptosis rate and blocked the cell in G2/M phase,but the effect on S phase is not obvious.conclusion:Intermediate Frequency Micro Current has certain inhibitory effect on U251 glioma cells.Intermediate Frequency Micro Current combined with elemene Injection can exhibit synergistic anti-tumor effect,the inhibition of glioma growth by inducing apoptosis and cell cycle arrest and apoptosis;It can cooperate with temozolomide to promote cell apoptosis and block U251 cells in G2/M phase.Through the study of intermediate frequency micro current,we can improve the efficacy and drug resistance of temozolomide in glioma.The research on the anti-tumor effect of intermediate frequency micro current provides a new idea for the treatment of glioma,which opens up a new direction for our independent innovation and development of new tumor physical therapy technology.At the same time,its action mechanism needs further exploration. |
