| Object:1.To observe the effects of different concentrations of adriamycin on the biological activity of H9C2 cells.2.The effects of curcumin on H9C2 myocardial cells induced by adriamycin biological activity,oxidative stress and apoptosis Impact of death.3.To investigate the protective effect of curcumin on adriamycin induced H9C2 myocardial cells.Methods:1.H9C2 cultured myocardial cells in vitro,intervened the cell culture for 12 h respectively with different concentrations of curcumin(5,10,20,40,80,160 μmol/L)and adriamycin(1,2,4,8,16,32 μmol/L),and detected the cell survival rate by MTT method.2.Experiment group:(1)normal control group: 12 h.(2)adriamycin group: adriamycin treated 12 h cells 8 μmol/L.(3)low dose curcumin group: Curcumin treated cells for 12 h to 10 μmol/L and 8 μmol/L doxorubicin.(4)common dose group in curcumin treated cells for 12 h to 20 mol/L curcumin and 8 μmol/L adriamycin.(5)the high dose group: Curcumin treated cells for 12 h to 40 μmol/L curcumin and 8 μmol/L adriamycin.3.(1)H9C2 myocardial cells were added to the corresponding drugs after 12 h growth was measured by MTT;(2)lactate dehydrogenase(LDH)cytotoxicity detection kit,lipid oxidation(MDA)assay measured detection kit and SOD activity detection kit LDH,MDA level and SOD activity.(3)Caspase-3,Bax and Bcl-2 protein levels were detected by Western blot method.Results:1.there was no significant difference between the different concentrations of curcumin in the growth of H9C2 myocardial cells by MTT assay.Curcumin had no obvious inhibitory effect on the proliferation of H9C2 cells(P>0.05).Compared with the control group,the concentration of adriamycin group can inhibit the growth of H9C2 cells(P<0.05),and the inhibitory effect of doxorubicin on concentration from low to high on the growth of H9C2 cells increased gradually,and the difference is obvious(P<0.05).2.the MTT test results showed that compared with normal control group,adriamycin group significantly decreased cell viability(P<0.05);compared with curcumin is low,middle and high dose group and adriamycin group,the cell survival rate was significantly increased(P<0.05).3.compared with the normal control group,the levels of LDH and MDA were significantly increased and the activity of SOD decreased significantly(P<0.05).Compared with adriamycin group,curcumin is low,medium and high dose group of LDH cells significantly decreased(P<0.05);compared with adriamycin group,the level of SOD cells in curcumin and high dose group increased significantly(P<0.01),the level of MDA was significantly decreased(P<0.01)4.Western blot detection results were as follows: Compa-red with the normal control elevated levels of Bax protein group,adriamycin group and Caspase-3 cells,reducing the activity of Bcl-2 protein(P<0.05);and compared with adriamycin group,curcumin is low,medium and high dose group of Caspase-3 cells,reduce the level of Bax protein(P<0.05),elevated levels of Bcl-2 protein(P<0.05).Conclusions:1.Adriamycin can induce the damage of H9C2 myocardial cells,curcumin has no obvious proliferation or inhibition effect on H9C2 myocardial cells.2.Curcumin has protective effects on adriamycin induced myocardial injury in H9C2 cells,which may play an important role in reducing oxidative stress and reducing apoptosis. |
