Pseudomonas syringae pv.syringae Vane Hall 1336 are important plant pathogens,sending major wide range,against a variety of woody and herbaceous branches,leaves,flowers and fruit,causing a variety of crop diseases,and the toxicity is related to T1SS,T3SS effector proteins.Pseudomonas syringae pv.syringae Vall Hall 1336 could synthesis phosphatidyl choline by pcs-dependent pathway which is depend on choline.According to the bacterial genome database,over 10%of the total bacterial synthesize PC,and the vast majority of PC-containing bacteria are associated with eukaryotic pathogens or commensal bacteria.The members of the laboratory have researched the action of phosphatidylcholine synthase gene(PCS)in bacterial pathogenicity,after more than two years,we have successfully constructed three type bacteria which are pcs mutants,and found that PC deletion causes Pseudomonas syringae pv.syringae pathogenicity lost;and the loss of pathogenicity are closely related to the functions of T1SS and T3SS.So after establishing functional PC-control T1SS and T3SS,further in experimental strains of wild type and pcs-mutant by comparing the major subunit assembly on cell membranes,the impact of the phospholipid component to protein topology,as well as gene transcription and expression to prove PC affect the mechanism of the secretion system function.This experiment by using the method of reverse transcription PCR to Analysis of 17 related genes of T1SS,34 related genes of T3SS Transcription in Pseudomonas syringe pv.syringe Van Hall 1336 and Pseudomonas syringe pv.syringe Van Hall 1336-pcs-clove pseudomonas wild type and not PCS for synthesis of PC-1336 of the mutant transcription,found on the Transcriptional horizontal,wild type and the mutant type 17 related genes of T1SS,34 related genes of T3SS could not show obvious differences.The results show that the loss of PC does not affect I,III secretion system related gene expression at the transcriptional level.
