In recent decades,the use of organophosphorus(OP)pesticides is one of the most important factors contributing to the increase in food production.Concomitantly,the presence of pesticide residue on foods such as vegetables and fruits is inevitable.OP compounds belong to a class of highly toxic neurotoxins that inhibit acetylcholine esterase(AchE)in the central nervous system synapses,leading to a subsequent loss of nerve function and to eventual death.Therefore,methods of degradation especially biodegradation is essential.Organophosphorus hydrolase(OPH)OPHC2 from Pseudomonas pseudoalcaligenes was successfully secretory expressed by Yarrowia lipolytica Polg and purified by using Ni2+ affinity chromatography in the present study.The purified enzyme was optimally active at 65 ? and pH 8.5.In addition,the purified enzyme displayed good thermal and pH stability using methyl parathion(O,O-dimethyl-O-4-p-nitrophenyl phosphorothioate)as a substrate.Moreover,Y.lipolytica is considered non-pathogenic;several processes based on Y.lipolytica have been classified as GRAS by the Food and Drug Administration(FDA).The cell culture supernatant can be used directly on vegetables and fruits that are contaminated by organophosphorus pesticides.
