The purpose of this study is that artificial synthesize ectomycorrhiza of Castanea mollissima and two kinds of ectomycorrhiza fungi,Lactarius hatsudake and Suillus granulatus.1.L.hatsudake and S.granulatus isolation were obtained from fruit body tissues,and maintenance of fungi in pure cultures,morphological characters of mycelium were observed in detail.The identity of the fungus in the petri dish was confirmed by the rDNA-ITS sequencing analysis.2.This paper studied on the optimum carbonic sources,nitrogenous sources,inorganic salts and the effect of growth factor on the growth rate of L.hatsudake and S.granulatus mycelium was studied by single factor experiment.3.Analysed the effect of different factor on the synthetic ectomycorrhiza.The number of mycorrhizal infection and the survival rate were contrasted when inoculated L.hatsudake mycelium suspension on the different age C.mollissima seedlings three months later.The test results demonstrated that the value of number of annual seedlings was higher than the germinated seedlings much more.According to the number of mycorrhizal infection judged which type of inoculum was optimum,it showed that the best inoculum type is mycelium suspension when the inoculum is L.hatsudake.4.The morphological characters of C.mollissima fresh ectomycorrhiza was observed and described systematically,in the cross section of the innoculated roots,the hignet was found,which confirmed there formed ectomycorrhiza in the roots.The identity of the colonizing fungus in the mycorrhizae synthesized was confirmed by the rDNA-ITS sequencing analysis.The two kinds identify methods improved the accuracy of the experiment.These results suggest that L.hatsudake and S.granula S.have a perennial ectomycorrhizal association with C.mollissima.The data provided in this work can be referred directly by the cultivation of mycorrhizal seedlings.
