The model of transformation of baicalin to baicalein by human intestinal bacteria in vitro was studied in this paper.The culture condition and methods of transformation and extraction were optimized,the key flora in the transformation were explored on the basis of the model had been established in our laboratory.First,the composition of the intestinal microflora in different groups were analysized and the key flora were determined according to the conversion efficiency of baicalin by intestinal bacteria of different individuals.Intestinal bacteria of different ages and different health status of population were selected to transform baicalin.The results showed that the bifidobacteria and lactobacillus may be efficient in the transformation of baicalin,and excessive E.coli.were inhibition to the transformation.Secondly,the culture and transformation method and solvent extraction were discussed.The dynamic cells and static cells were used to transform baicalin,the dynamic cells was better.The ultrasonic treatment and Triton X-100 treatment can be adopted in breaking the cellwall,the Triton X-100 was better.The different solvents: methanol,ethanol,aether,ethyl acetate and acetone were discussed in the experiment,the results showed ethanol was the best.Finally,the culture conditions were optimized.The influence of carbon source on transformation was investigated;The optimum time,temperature,pH and solid-liquid ratio were determined by the orthogonal experiment of four factors and three levels.As a results,the optimum culture conditions were as follows: the carbon source was inulin,the pH of the culture medium was 7.2,temperature was 48?,time was 33h,solid-liquid ratio was 2:100.
