The research,complete mitochondrial genome were identified by the High-Throughput Sequencing and the Polymerase Chain Reaetion sequeneing for 4 genera and 5 species of the Ponerinae.Which are Anochetus yunnanensis,Cryptopone sauteri,Pachycondyla luteipes,Anochetus risii and Ectomomyrmex sp.The above content for Complete Mitogenomes of the 5 species were properly analyzed and annotated.Associated with the Formicidae whole mitochondrial genomes which from the NCBI has issued.The monophyly of the 5 species of ants is tested through phylogenetic analysis based on data combined with mitochondrial genomes,apply to the coxl gene,cox2 gene,rrnL gene,rrnS gene,cox]+cox2+rrnL+rrnS gene and PCGs combined data.The study has practical value and theoretical significance in Evolutionary Genomics for Molecular Evolution and Molecular Systematics.The conclusions drawn from the study are as follows:1.The complete mitochondrial genomes of Anochetus yunnanensis,Cryptopone sauteri,Pachycondyla luteipes,Anochetus risii and Ectomomyrmex sp.are 14962bp,14988bp,15246bp,15556bp and 15146bp in size respectively.The mitochondrial sequence consists of double-stranded DNA molecule,including 13 protein-coding genes,2 ribosomal subunit genes,22 transfer RNAs,and a non-coding AT-rich region,which is nake up with 2 strand.The genome arrangement is typical of insect taxa and make consistency to the Ponerinae has published.2.The base composition of the 5 species has obvious bias with AT,The A+T Content were 86%,86%,84.2%,82.3%and 86.2%,respectively,in that AT content of the protein-coding genes 84.8%,84.9%,82.5%,80.4%and 80.9%,respectively,which the AT content of third coxdon position is the rnost.3.The starting codon of Anochetus yunnanensis,Cryptopone sauleri,Pachycondyla luteipes,Anochetus risii and Ectomomyrmex sp has general start with ATN,TAA is the most frequently used termination codon.The amino acid has been composed by the mitochondrial protein-coding genes.The number of codons UUU,UUA,CUU,CUA,AUU and AUA is the dominant,which is rich in A and T codons.That means,the codons has been proved the AT bias of whole mitochondrial sequence.4.There are 22 tRNAs of the 5 species of the Ponerinae,and the length is between 59-73bp.It can be folded to form a clo'ver structure.It contains an amino acid acceptor arm,a DHU stem and a DHU ring,an anti-codon stem and an anti-codon ring,a T?C stem and a T?C ring,and a variable ring.tRNA-ser and tRNA-leu are the unusual tRNA which above all.They have double brother genes:tRNA-serUCN,tRNA-serAGN,tRNA-leuUUR,tRNA-leuCUN.5.We have token the coxl gene,cox2 gene,rrnL gene,rrnS gene,cox1+cox2+rrnL+rrnS gene and PCGs combined with the ants data into phylogenetic analysis.At last the 5 species of the Ponerinae who can be clustered together,based in the Phylogenetic analysis tree,Myrnicinae,Formicinae and Dolichoderinae clusterd with alone group.
