Zika virus belongs to the Flavivirus.It is transmitted by aedes mosquitoes,and will causes some mild symptoms including headache,fever,muscle pain and joint fatigue.For a long time,zika virus was not considered a threat to public safety.However,since May 2015,in South America,an outbreak of zika virus infection happened and suggested that neonatal microcephaly and adult guillain-barre syndrome and other serious diseases was related to zika virus infection.Outbreaks caused by the damage alarming,zika virus outbreak caused widespread concern.The world health organization(WHO)announced zika virus outbreak of public health emergencies.Zika virus infection is regarded as a major threat to human health threat.Unfortunately,at present there is no drug special to zika virus infection or vaccine for prevention.Zika virus nonstructural protein NS3 has two domains,for N terminal the protease domain and C terminal the helicase domain.The protease domain cuts polymer precursor protein to form mature viral protein function,while the helicase domain unwinds the genome in viral replication after forming double-stranded RNA.NS2 B protein has the effect of stabilizing NS3.Therefore,NS2B-NS3 protein plays an important role in the process of virus replication.Studies shows that in falvivirus NS3 can serve as antiviral drug targets.Reseraches into NS3 can promote understanding of zika virus and have important significance on drug development.This article,firstly,in the protease active study of NS2B-NS3 protein,compared with the activity of protease domain,we established that full length NS2B-NS3 protein has better activity.Secondly in the inhibition of serine protease inhibitor aprotinin experiment,we proved that the state of the total length of NS2B-NS3 protein have better sensitivity to inhibitors.Finally,we screened a species-specific inhibitors.From what has been discussed above,in the zika virus nonstructural protein NS2B-NS3 enzymology and inhibitor screening tests,we deepened the understanding of the zika virus,and provided the direction for specific treatment drug research of the virus infection.
