Polyvinyl alcohol(PVA),which is polyvinyl alcohol,is a highly efficient water-soluble polymer that is also commonly seen as a vinyl alcohol monomer.This biomacromolecule has good adhesiveness and is generally used in the industrial papermaking and textile fields because it is difficult to be degraded and becomes one of the pollutants that affect the environment.Therefore,the use of biological proteases to decompose green PVA is convenient and the reaction speed is fast.It can also reduce the energy consumption and cost in the original method to a great extent,and it can also reduce the pollution of polyvinyl alcohol to the environment.However,so far,the source of PVA degrading enzyme production is very low and production technology is weak,and basically comes from microbes.Therefore,the more extensive sources of PVA degrading enzymes,more efficient degradation rates,and the study of metabolic processes that occur after degradation,have very broad prospects and implicationsFirst of all,we found a polymer in the MALDI analysis of the plant sample solution.We analyzed and identified this polymer as polyvinyl alcohol,PVA.Then we discovered this polymer in different tissue parts of Arabidopsis and found that PVA has a very important role in the growth and development of Arabidopsis thaliana.After bioinformatics comparison and analysis,it was found that its degrading enzymes all contained a PQQ domain.Therefore,we found the degrading enzyme AtAEE19 containing this domain in Arabidopsis thaliana.PVADH(PVH1,PVH2,PVH3)containing this domain was found in Cotton.We constructed expression vectors for the degradative enzymes such as PVH1,PVH2,and PVH3 in cotton,and also constructed vectors for AtAEE19,exploring the degradation and degradation activities of PVA.We used a method of constructing an electron transfer chain to measure the degradative enzyme activity by measuring the reduction of DCPIP at 1600 using a 1 cm path in a spectrophotometer.Experiments showed that degradation enzymes AtAEE 19 and PVADH were able to degrade PVA and detect the activity of PVA degrading enzymes.PVA is degraded as a substrate.After degradation,small molecule metabolites can be generated.How do metabolites affect plants?We identified the product after degradation of PVA and found that AtAEE 19 degraded PVA to produce a 3-hydroxypropionic acid substance that would enter the body and affect plant growth and development.In order to verify the above biochemical results,we used PVA and 3-hydroxypropionic acid to treat Arabidopsis thaliana,respectively,and found that both had similar functions and phenotypes,they can affect seed germination and lateral root growth.Therefore,PVA degrading enzymes can degrade PVA to produce degrading enzyme activity.After degrading PVA,metabolite 3-hydroxypropionic acid is generated and entered plant cells,which affected the development of Arabidopsis,especially the germination of seeds and the growth of lateral roots.
